Chronic Renal Failure Animal Model Preparation Method And Rhubarb Anthraquinones Of The Pharmacodynamic Study

Objective:We designed five SD rat models of chronic renal failure(CRF), which is adenine administrated,3/4 nephrectomy,5/6 nephrectomy,ligation the up and down poles of unilateral kidney,ligation the up and down poles of unilateral kidney and refrigeration the outside cortex of the same kidney.Contrasted to the five rat models,we study their stability.And we preliminary investigate the potency of nomadic anthraquinone of Rhubarb to the CRF.Methods:SD rats were randomly divided into six groups:one normal group and five model groups,modelⅠgroup(adenine lavaged),modelⅡgroup(3/4 nephrectomy),modelⅢgroup(5/6 nephrectomy),modelⅣgroup(ligation the up and down poles of unilateral kidney),modelⅤgroup(ligation the up and down poles of unilateral kidney and refrigeration the outside cortex of the same kidney).Four and eight weeks after procedure,we observed general condition and weight and mortality of rats,the urea nitrogen and creatinine content of rat serum.We collected the 5/6 nephrectomy CRF model and administrated nomadic anthraquinone of Rhubarb to the model rats in order to observe general condition and weight of rats,the urea nitrogen and creatinine content of serum,and the pathological change of residual nephridium.Results:We compared with the model groups and the normal group,1.Rat weight of model groups decreased(P＜0.05);2.The urea nitrogen and the creatinine content of model rat serum raised(P＜0.01);3.After four-eight weeks procedure, rats mortality of modelⅢgroup reduced than other model groups;4.We compared with the treatment groups and the model group after four weeks administrattion with nomadic anthraquinone of Rhubarb.In treatment groups,the urea nitrogen and creatinine content of rat serum were decreased(P＜0.01或0.05);the number of renal tubules and glomerulus were increased(P＜0.05);the average area and perimeter,maximum and minimum diameter of glomerulus were decreased(P＜0.01或0.05);the average area of basilar membrane and mesangial cells of glomerulus and their light density were reduced.(P＜0.05).Conclusion:Contrast to the other rats models of CRF,5/6 nephrectomy is a stable and perfect CRF model.Nomadic anthraquinone of Rhubarb can decrease the urea nitrogen and the creatinine content of CRF rat serum,inhibit the fibrosis and embolization of glomerulus,improve the sclerosis of renal tubules,restrain the accrementition of frill and delay the progress of CRF rats.
