Xin Hot Drugs On Normal And Deficiency Rat Model Of Hepatic Metabolic Enzyme Cyp3a Activity And Protein Expression

Objective:To observe the drugs impact to the activity of CYP3A of hepatic metabo -lic enzymes by using the effect of drugs and monomer composition to normal rat liv -er microsomes,by comparison of three types of different rats:normal ones,deficien -cy-cold ones and the rats with drugs,studying the difference in the activity of CYP3 -A and the change in protein expression to inspect the correlation of drugs,model an -d the metabolic enzymes.Methods:To measure the activity of CYP3A in normal rats and Hydrocortisone intr -amuscular injection in the replicated model of Deficient-Cold rats by using Calcium chloride precipitation to prepare hepatic microsomes;the total protein was prepared with RIPA and to measure the protein expression of CYP3A using Western Blot.Results:when inspecting the activity effect of CYP3A caused by some drugs we fou -nd that there are different levels of induction with the crude extract which at low co -ncentrations from xianmao,fuzi and huangbai separately,fuzi and huangbai induced significant difference at the concentration of 0.25,0.50mg/mL,and xianmao has a very significant difference in inhibiting effect of CYP3A.Monomer composition hyp - aconitine whose concentration was 2.5μg/ml has the strongest induction,but it also showed a certain degree of inhibition at the concentration of 25μg/ml;Aconitine indu -ction to CYP3A increased with the concentration accumulation;The role is of comp -arative ease Berberine.The activity of CYP3A is variant among three different organic conditions normal rats,model rats and model rats with drugs:the activity of CYP3A in liver of Deficient Cold Condition group is significantly lower than that of Normal Condition group;its activity in Drug group is higher than models rats group.The protein expression of CYP3A among four groups has no significant differences.Conclusions:The tendency of drug crude extracts to the activity of CYP3A is the similar as the corresponding monomer;the activity of CYP3A is variant among three different organic conditions-normal rats,model rats and model rats with drugs, however,the protein expression of CYP3A has no variance.
