Aerobic Exercise And Dietary Control Of Type 2 Diabetes Mellitus Rats Highest Quintile Of Adiponectin, Ampk, Glut-4 Influence

Objective:To explore the effect of aerobic exercise training and dietary control on Adiponectin-AMPK-GLUT-4 signaling pathway in type 2 diabetic rats and its interaction.Method:62 male SD rats were 6-week-old, 8 rats were randomly selected as normal control group (Control, C group), which fed with normal standard diet. The remaining 54 SD rats were feed high-fat and high-carbohydrate diet for 6 weeks and then were injected low-dose streptozotocin though abdominal cavity to establish animal models of type 2 diabetes. and then the rats with type 2 diabetes were randomly divided into 4 groups : DM control group (DM,n=9),DM+ aerobic exercise training group (DME,n=10), DM+ Diet control group (DMD,n=10), DM+ aerobic exercise training + Diet control group (DMED , n=10). The rats in DM group were continued to feed high-fat and high-charbohydrate diet and did not exercise training. Exercise Training used 60 minutes'unload swimming, 6 times per week. The dietary control used the standard diet with the same dose in the DM group. 13 weeks later, FPG, FINS, plasma Adiponectin,musclar AMPK and GLUT-4 concentrations in every group's rats were examined.Results:(1)After 13 weeks of the establishment of type 2 diabetes model, compared with C group, the FPG contents increased significantly in DM group(P﹤0.01), the serum insulin concentration in DM group decreased significantly(P﹤0.01). GLUT-4 and AMPK concentration in skeletal muscle significantly decreased (P<0.01), muscle glycogen concentration was no significant changes (P﹥0.05), serum adiponectin concentration decreased significantly in DM group(P<0.05).(2)Through the double factor variance analysis, aerobic exercise training for 13 weeks could make the FPG contents significantly to decreaser(P﹤0.05). Despite FINS and skeletal muscle GLUT-4 concentration had some increase, but there were no significant difference (P> 0.05), and muscle glycogen content increased significantly (P <0.05), serum adiponectin and skeletal muscle AMPK was significantly increased (P <0.05).(3)Through the double factor variance analysis, there was not significantly affect of 13 weeks of the dietary control on FPG levels in diabetic rats (P> 0.05), but significantly increased FINS level (P <0.05). Muscle glycogen, GLUT-4 and AMPK content in skeletal muscle did not change significantly (P> 0.05).(4)Through the double factor variance analysis, there were no significant interactions of 13 weeks of aerobic exercise training united dietary control on FPG, FINS, adiponectin (P> 0.05), there was significant interaction on GLUT-4 and AMPK content in skeletal muscle (P <0.05), However, there was not significant interaction on muscle glycogen content in the diabetic rats(P> 0.05).Conclusion:(1)Establishing of the Model of type 2 diabetes in this research was successful. At the same time, adiponectin-AMPK-GLUT-4 signaling pathway in diabetic rats blocked, which made skeletal muscle to transport glucose decrease, blood glucose concentrations increased.(2)13 weeks of aerobic exercise training could increase the ability of insulin secretion of pancreaticβ-cells, which significantly reduce blood glucose in diabetic rats. In addition, to some extent, 13-week aerobic exercise training could improve adiponectin-AMPK-GLUT-4 signaling pathway in diabetic rats to enhance the ability of skeletal muscle to uptake glucose from the blood.(3)In the treatment of diabetes, relying on dietary control simply and could not effectively reduced the FPG. level in diabetic rats. And 13 weeks of dietary control improved the adiponectin-AMPK-GLUT-4 signal pathway in diabetic rats was very limited. (4)There were no significant interaction of dietary control united aerobic exercise training on FPG levels and improving the adiponectin-AMPK-GLUT-4 signals pathway in diabetic rats...
