Objective: to explore the therapeutic effect of traditional Chinese drug on nonalcoholic fatty liver rats chronically fed with high-fat and drug diet.Methods:44 Wistar rats were induced to the model of NAFLD with high fat and drug diet that was made up based animal feed(77%), lard(10%), vitellinepowder(5%), sucrose(5%), cholesterol(2%), bile salt(0.5%), and thyreostat(0.5%).after 6 weeks , randomly selected 6 rats , Executed and free liver, observed under the microscope , see cytoplasmic filled with a large number of lipid droplets of varying sizes, confirming the successful model .except the model group, the group of traditional Chinese drug, Simvastatin group and tiopronin group were treated by corresponding medicine. After 4 weeks, all rats were put to death, and the following items were detected:the liver index through measuring the body weight and weight of liver the serum levels of ALT,AST;the serum contents of TC,TG and leptin, tumor necrosis factor (TNF-α), blood glucose(GLU), insulin(INS),and to calculate the insulin resistant index(HOMA-IR) with them.the contents of malondialdehyde(MDA) and the activity of superoxide dismutase(SOD)were detected in the liver tissue homogenate.The liver histopathology was observed by hematoxylin and eosin(HE)staining. Results: After to analyze the experimental data,we found that, the indexes are all improved in treated groups than those in model group(P<0.05). The indexes of GLU,INS,MDA,SOD are all improved better in traditional Chinese drug than those in Simvastatin group and tiopronin group(P<0.05),and for others, traditional Chinese drug is not better than Tiopronin group and tiopronin group, But its up Simvastatin a single lipid-lowering and Tiopronin the protection of the liver, the treatment must have advantages.Conclusion:There are several mechanisms on traditional Chinese drug preventing and curing NAFLD.①the traditional Chinese medicine can treatment of experimental nonalcoholic fatty liver with a certain degree, its mechanism may be related to regulation of fat metabolism, improve liver function, antioxidant effects;②microscopic mechanism of inhibition may be related to insulin and leptin resistance, reduce release of tumor necrosis factor. |
