Yiqihuoxue Drug Glomerular Mesangial Cell Matrix Metalloproteinase Protease-2 And Its Inhibitor -2

Purpose: To study the effect of huang qi and tai zi shen, dan pi and dan shen, zi cao and yi mu cao on matrix metalloproteinase-2,tissue inhibitor of metalloproteinase-2 and m-RNA expression of them, investigate every group herb in preventing mesangial selerosis.Material and method: 1.Through the phamacology of serum , we got every group herb's harmaeologieal serum. 2. Culture the glomerular mesangial cells and prolifelate them by AngiotensinⅡ. 3.m-RNA pression of MMP-2,TIMP-2 were measured by RT-PCR. 4.The secretion of fibronection(FN) and collagenⅣexamed by enzyme—linked immunosorbent assay(ELISA).Results:1. AngiotensinⅡincreased the secretion of FN and collagenⅣ,compared with 10% FBS there is obviously difference (p<0.05) .2. Every group herb didn't effect on MMP-2.3. Huang qi and tai zi shen obviously inhibited the expression of TIMP-2, compared with pathology group there is obviously difference (p<0.05) . Other groups also could inhibite the expression of TIMP-2 but are not remarkably.Conclusion:1. Huang qi and tai zi shen obviously inhibited the expression of tissue inhibitor of metalloproteinase-2 m-RNA.2. Huang qi and tai zi shen can prevention and treatment of glomeruselerosis by inhibiting the expression of tissue inhibitor of metalloproteinase-2 m-RNA.3.There is the tendency to inhibiting the expression of tissue inhibitor of metalloproteinase-2 m-RNA, which was made by dan shen and dan pi,yi mu cao and zi cao.
