γ-ray, also known asγparticles. It was a kind of electromagnetic radiation produced by the interaction of subatomic particles and the wave length is shorter than 0.2 angstrom.γ-ray has a strong penetrating power, the energy is higher than 100 keV. A large number of studies have shown that radiation can cause great changes on animal life cycle, physiological characteristics, especially in the aspects of insect breeding and survival.It has been approved thatγ-ray has a big effect on lifespan, fecundity, activity and some other aspects during the development of kinds of animals. What's more it also has advantages in tumour cure and immunity strengthen in some cases. At present, national researches about the effects of radiation are mostly focused on the pest control, which is used to decrease the numbers of the progeny or to produce some sterile ones. At the mean time, foreign researchers pay more attention to the biology effects of the radiation, especially the function to induce the genome instability and chromosome aberrance, and the radio-induced hormesis and radioadaptation. Furthermore most of the reports are concentrated on the effects of radiation during the earlier developmental phases, but the radiation effects on the adult ones are seldom reported. Basiced on the characters of fruit fly Drosophila melanogaster and used to be an ideal model organism for studying ageing in biology. What's more there are many genes, basic signal pathway and biochemical processes are highly conserved in human and Drosophila. Therefore we confined our attention to the effects and mechanism on aging, activity, endurance, the morphological changes of the brain and eyes and some other aspects as well as the relationship among them after the adult flies were irradiated by theγ-rays.Here, we given two doses of 50 Gy and 100 Gy to the adult male flies with different developmental phase, and studied the survivorship, medium life span, maximum life span, locomotor activity, starvation and heat shock endurance of the irradiated males flies, and the fresh body weight, the height of the pupa of the F1 progeny, the morphological changes of the brain and eyes of the F1 flies, and the changes of apoptosis related gene Grim. At last we analysed the effects and mechanisms of irradiation with the male flies on aging and development as well as the relativity among them. The results showed:1.γ-ray significantly reduced the middle lifespan, mean lifespan, maximum lifespan and the 90% survival rate of the adult flies irradiated with different developmental phase.2. Irradiation decreased the resistance to both starvation and heat shock. And the locomotor activity were lower than the control at the beginning of life, but it was recovered later and has significant difference with the control.3. Radiation reduced the level of SOD expression and accelerated the progress of aging, and it has the similar trend with the decrease of lifespan.4. Radiation with the parent male flies enhanced the body weight of the F1 progeny, and the higher dose ofγ-ray can raise the height of the pupa of the elder male flies' progeny.5.γ-ray also has effects on the morphological changes of the brain and eyes of the F1 flies.6. Different dose ofγ-ray on male flies makes 3 base pair changed in CDS of grim gene and 19 bp lost from the 1139 site to 1211 site, and the correspond anmio acid sequence is LLQLLQ. |
