| Purpose: The study aims at studying the effect of Astragalus extract on theskin of natural aging mouse in hydrogen peroxide (H2O2) content, hydroxyproline (HYP) content, as well as the ultrastructural changes of the skin, to detect the function of Astragalus extract in delaying the role of natural aging mouse in skin.Material and method: 12 female Kunming mice of 4-month-old, weight of 27~34 gram, as the young control group(A), 60 of female Kunming mice of 18-month-old, weight of 48~55 gram, as the old age group, and then were randomly divided into 5 groups , with every group having 12 Kunming mice of 18-month-old : elderly control group(B), low-dose group of Astragalus (C), middle dose group of Astragalus (D), high-dose group of Astragalus (E), positive control group of older(F). In accordance with sub-groups feeding cage. Group A and B were administered0. 9% normal saline (NS) 20ml/kg, group C, D, E were irrigated respectively with Astragalus extract of 4g/kg, 8g/kg, 16g/kg. Group F were administered with vitamin E-C suspensi of 3mg/kg. Going on 30 days, once a day. The first 31 days all the mice were executed and derived.Adopt technology pathological tissue, HE stain, 40 times of light microscope to observe the structural changes in the dermis, and to determine the content of HYP and H2O2 by biochemical analysis.Results:1. Compared with group A, there is significant increase in content of H2O2 , in skin of organizational structure of Group B, C, D, E and F(respectively is: p<0.01, p<0.01, p <0.05, p<0.05, p<0.05) ; that HYP contentsreduces (respectively p<0.05, p<0.05, p <0.05, p<0.01, p<0.01).2. the skin biopsy of five elderly mice showed a state of natural aging. Compared with the elderly control group(B), the H2O2 content of the elderly RA group (C, D, E) and elderly positive control group (F) reduced significantly 2. respectively is : p<0.01, p<0.01, p<0.05, p<0.05) ; the content of HYP increased significantly( respectively is p<0.05, p<0. 05, p<0. 01, p<0. 05).Conclusion:1. In the skin tissue of natural aging mice, the hydrogen peroxide content was increased, and the hydroxyproline content was decreased.2. Astragalus extract can reduce the content of hydrogen peroxide and increase the content of hydroxyproline in skin tissue of natural aging mice to delay skin aging. |
