Ginger is the fresh rhizome of Zingiberaceae. Modern study found that ginger have various biological effectiveness,such as oxidation resistance,free radical scavenging,cholesterol lowering, antiatheroscloresis,liver and gallbladder-protection and etc.Then,the free radical from the alcohol metabolism is one of the important factors that cause lipid peroxidation and alcoholic liver injury.So ginger have huge development potentials in the prevention and treatment of alcoholic liver injury because it has the effect of oxidation resistance and liver-protection.This research want to study the ethanolic extract from ginger for the protection of the alcoholic liver injury,and separates the ethanolic extract for the test of oxidation,then selects the best active region.1.The preparation and separation of ginger ethanolic extract.The ginger ethanolic extract is getted by ethanol lixiviation, decompression concentration, freeze drying. Then the ginger ethanolic extract(crude extract) is separated by AB-8 macroporous resin to water region,25% ethanol region,50% ethanol region,75% ethanol region,95% ethanol region.2.The test of gingerol in crude extract and separating product.The standard curve was drown according to the vanillin standard substance,gin- gerol in crude extract and separating product were tested.The gingerol content (6.4%)in crude extract was determined,while water region 1.3%,25% ethanol region 11.5%,50% ethanol region 34.7%,75% ethanol region 32.3%,and 95% ethanol region 28.5%.3.The test of anti-oxidation activity in crude extract and separating product.The lipid peroxidation model was made to produce MDA,the inhibition ratio of crude extract and separating product to MDA was tested. The 50% ethanol region is the best effect whose MDA IC50 is 0.3mg/mL,the MDA IC50 of 75% ethanol region is 0.5 mg/mL,while the activity of the both are better than VitE.The result of similar SOD (1.0mg/mL)showed that the SOD activity of 50% ethanol region and 75% ethanol region are 19.59 U/mL and respectively 19.05 U/mL, but no significant difference was found between the corresponding concentrations. 4.The protective effects of the ginger ethanolic extract to alcoholic liver injury on mice.The mice was divided into five groups: the normal group,the model group,and the low, middle, high doses group. Except for the normal group,every group was treated with 50% alcohol 10mL/kg bw i.g for alcoholic liver injury. Low,middle, high doses group was given 120mg/kg, 240mg/kg, 480mg/kg of the Ginger ethanolic extract for protective effects. Normal group was given the equivalent amount of distilled water for contrast. After being continuously treated with 30d, measure the ALT ,AST, MDA, SOD values of the mice from each group.The ALT,AST,MDA values in the model group mice was increased while the SOD values decreased,the significant difference was found when compared with that of the normal group(p<0.05或p<0.01).The indexes of the control group improved in different degrees comparing with model group. The protective to liver injury compared with the contral effects in the middle and high doses groups were very significant(p<0.01).
