Purpose:To explore the pathogenesis of irritable bowel syndrome (IBS) , and Tong- xieyaofang is used to research effect of Mast cell(MC), 5-hydroxytryptamine(5- HT) and Nervous growth factor(NGF) in the Colon of irritable bowel syndrome model.Material and method:The animal we choosed is neonatal SD rats, 8 days old, Body weight 10g±2g. Model rats of IBS were prepared by chronic stimulation in colon(two weeks) and stimulation on tails(7 days)to sets up the model of intestinal hypersensitivity and to evaluate model. After verification establishing model successful, rats were randomly divided into six groups: the addition of the normal control group (Group A),the model group(Group B),the low dosage of Tongxieyaofang group (Group C),the middle dosage of Tongxieyaofang group (Group D),the high dosage of Tongxieyaofang group (Group E), with the Spasmomen control group(Group F). Group A and B received oral administration in 1ml/100g,Group C,D,E received oral administration in 0.25ml/100g,0.5ml/100g,1ml/100g and Group C received oral administration in 1ml/100g,once everyday for a month. And then ,We should observe the bulgy capability of the sacculus that cause up lifting of abdomen and arching of back and the time of contract about abdomen muscle in rats.Toluidine blue staining by measuring statutory Organize content of mast cells, immunohisto- chemical method to observe 5-HT and NGF positive cells and expression.Results:1.After treatment, high dosage of Tongxieyaofang and Sibamin induced intestinal sensitized behavioral indexes returned to normal, Compared with Group B(P<0.05,P<0.01);In 0.3ml, 0.6ml, 0.9ml capacity expansion, abdominal muscle con- tractions were significantly decreased(P<0.01,P<0.05), and there is no significant difference between group E and F. 2. After treatment, group B density was significantly higher than group A( P<0.01) , irregular shape were common, some cells rupture, overflow membrane particles, and even the formation of vacuoles. group E,F compare with group B density was significantly reduced( P<0.01),shape is rule. Account for the high dosage of Tongxieyaofang and Sibamin can reduced the number of Mc and significantly inhibited degranulation phenomenon.3. After treatment, the group B immunostaining positive cells of 5-HT and NGF was significantly increased(P<0.01),and the expression increased significan- tly compared with the A , group E,F compare with group B the immunostaining positive cells of 5-HT and NGF was significantly reduced( P<0.05). Account for the high dosage of Tongxieyaofang and Sibamin can reduced number of 5-HT and NGF positive cells and tissue of expressionchemical expression.Conclusion:1. The high dose of TongxieYaofang was able to make intestinal behavioral and electrophysiological indicators induced after sensitization returned to normal.2. Tongxieyaofang can inhibit mast cell degranulation in rats, reducing the number of mast cells;and reducing the number of 5-HT and NGF immunostaining positive cells and expression.3.Tongxieyaofang can inhibit the release of MC activation and chemical tran- transmitters of 5-HT and NGF directly , or indirectly inhibit MC activation and sensitization mechanism to reduce the 5-HT and NGF Immunohistochemical expre- ssion,it cuts down the excitability of the dorsal horn nerve fible to raise the threshold value of visceral pain.So MC,5-HT and NGF play an important role in the pathophysiology of IBS, and may be involved in nerve - Immune - regulation of endocrine networks.
