The Preliminary Research Of Polysaccharide Of Codium Fragile On Bioactivity

Polysaccharides of Codium fragile were extracted by means of hot water extraction and ethanol precipitation, to study the mechanism of promoting tumor apoptosis, tumor inhibiting and tumor migration as well as inhibiting effects of polysaccharide of Codium fragile (CFPS) on blood coagulation of polysaccharide of Codium fragile were preliminary researched.MTT colorimetry was applied to quantify cell vitality of the cell lines A549 after being treated with polysaccharide of Codium fragile. Hoechst33258 dyeing A549 cells treated with polysaccharide of Codium fragile have been used to observe the morphological changes of apoptosis after the A549 cells treated by different concentration polysaccharide. The expression of apoptosis related proteins Bcl-2 of A549 cells treated by polysaccharide of Codium fragile with different concentration 24h are determined by Western blot and immunocy to chemistry. The migration ability of A549 cells treated by polysaccharide of Codium fragile was observed by means of Boyden chamber model.The blood plasma is made of blood,Which is extracted from New Zealand white rabbit's heart .we made the blood plasma mixed with the physiological saline and the polysaccharide of Codium fragile solution which density are 125,250,500 and 1000μg/ml.; rabbit blood activation thrombin time (APTT), prothrombin time (PT) and thrombin time (TT) was determinated by mixed blood condensing apparatus to know the influence of polysaccharide of Codium fragile on anticlotting effect. Carotid artery intubation extraction was applied to get New Zealand white rabbit blood then get rich platelet plasma (PRP) and lean platelet plasma (PPP); We mingled them into physiological saline and the polysaccharide of Codium fragile solution which density are 125,250,500 and 1000μg/ml, and taken ADP as exutory,though this method we can observed the maximum gathered rate.The results showed that A549 cells were inhibited obviously by CFPS which depend on the drug dosage and treat-time closely; It come to a conclusion that the tumour cells could be inhibited by CFPS which was positive correlation with treat-time of the CFPS; the expression of Bcl-2 in tumour cells treated by CFPS decreased and leaded to tumour cells apoptosis; the experimental results of Boyden chamber model showed that polysaccharide of Codium fragile can inhibit A549 cell migrating and reduced tumour cells permeating to peripheral tissue. APTT, PT and TT were prolonged significantly by polysaccharides and the platelet aggregation was inhibited prominently.The experimental proved that polysaccharide of Codium fragile can inhibit tumour cells proliferation, migration and inhibit blood coagulation and thrombosis.
