Because of the integration and miniaturization characteristics of microfluidic chip, the processing time can be greatly reduced, while the detection resolution and accuracy was increased, so that consumption and costs significantly reduced. Based on these benefits, microfluidic chips has been widely used in the fields of chemistry, bio-medical field, optics and information science and other fields. Microfluidic chip detection technology also plays a very important role in the process of continued utility of microfluidic chips. The microfluidic chip was most often used in area of life sciences, so its detection has special requirements.With the continuous development and enhanced of detection technology, the detection methods and techniques for microfluidic chip have been continuously improved, it is developing from optical detection, electrochemical detection, mass spectrometry and other detection methods gradually to Information Fusion direction.This paper, supported by Tianjin Natural Science Foundation"The research of microfluidic chip and detection system used in the measurement of intracellular calcium ion concentration", saw detecting intracellular calcium ion concentration of as the experimental basis to research a kind of microfluidic chip detection method based on image processing. In the experiment , fluorescence ratio method was used as the calculation basis to detect the concentration of calcium ion, combinied fluorescence ratio method detection principle and image information processing to analysis the experimental data,so as to provide accurate parameters for the calculation of calcium ion concentration.The main contents of this paper include:1,Researched a kind of microfluidic chip which was used in vivo cell culture, cell detection, reagent feed integrated, designed a microfluidic chip and the analysis integrated system for the detection of intracellular calcium ion concentration.2,The study of existing microfluidic chip detection methods, image processing will be applied in the detection system. 3,Studyed the principle of ratio fluorescence method to detecting intracellular calcium ion concentration,designed a hardware system for experiment.4,Researched microscopic fluorescence image processing methods, analysis and processing of the cell image obtained from experiments.This paper designed a microfluidic microscopic imaging system, xenon lamp was used as the excitation source of this system, the main components are microfluidic chip-driven system, fast wavelength switching system, micro system and imaging systems and so on.In the detection,this system used software to obtain real-time cell fluorescence images and displayed, processed and analyzed them,then the fluorescence position and intensity was extracted to provide experimental basis for intracellular calcium ion concentration,finaly,a kind of microfluidic chip detection method based on image processing was determined. The study of this detection method provided theoretical and experimental experience for the development of microfluidic chip detection methods, also opened a new chapter for microfluidic chip in the field of cell analysis.
