Anisaldehyde is an important organic intermediates, it has a lasting aroma of hawthorn and strong antioxidant properties, widely used in food, medicine and daily chemical industries. Currently anisaldehyde products mostly prepared by chemical, has serious drawbacks. The newly developed electro-oxidation process and ozonation process, although in environmental issues has been greatly improved, but harsh reaction conditions, equipment for high, industrial difficulties. Therefore, to find a cost-effective, environmentally friendly way to anisaldehyde preparation is worth studying.With trans-anethole as the only carbon source,320 strains were isolated and enrichmented from 50 anise environmental samples, further screening resulted in a strain WGB30 by Spectrophotometry,Thin-Layer Chromatography and Gas Chromatography,with higher productivity of anisaldehyde,Moore generation rate of anisaldehyde is up to 5.08%. According to the morphological features, the results of physiological and biochemical experiments,Biolog EcoPlates identification and 16SrDNA sequence analysis, this strain was identified as an organism of the genus Burkholderia sp.To improve anisaldehyde production of strain WGB30,this test has studied into main components of medium, culture conditions,including initial pH, temperature, medium volume, wave bed speed and transformation time, seeds growth time, biomass, emulsifier and the add time and additives of trans-anethole. By single factor experiment and orthogonal experiment,the optimal conditions of strain WGB30 were as followed:glucose 20g/L, peptone 8g/L, initial pH6.5,the fermentation temperature 28℃,250ml flask containing 60ml, speed 180rmp, the transformation time 3d.In the above fermentation conditions, Moore generation rate of anisaldehyde can reach up to 11.42%, which is about 2 times compared with before optimization.The metabolites of Burkholderia WGB30 bioconversion trans-anethole into anisaldehyde were analysised by Gas Chromatography-Mass Spectrometry (GC-MS), detected three products,anisaldehyde, anisic acid and N-methyl-ptoluidine,respectively.And preliminary estimated metabolic pathways of Burkholderia WGB30 biotransformation trans-anethole.
