Morus.nigra Linn on the plant taxonomy is the Moraceae,mulberry genera, black mulberry varieties. Mainly distributed in Akesu, Hetian and Kashgar and other regions,is the only flower of chromosome ploidy in multiples of 22 mulberry varieties, but also classification and identification of mulberry varieties in Xinjiang is currently the only species of the black mulberry (N.nigra Linn) in the morphological classification as a separate species, and is a primitive type.Xinjiang is the only possession and cultivation of Morus.nigra Linn appropriate areas, has a unique natural environment and significant.The geographical advantage to develop Morus.nigra Linn as raw materials and herbal agentshas a bright future.Polysaccharide is extracted using the methods of hot water extraction and ethanol precipition in this study.The content of polysaccharide was measured by phenol-sulfuric acid method.Morusnigra Linn polysaccharide use Sevag method to remove protein.Polysaccharide dissolve, adding an appropriate amount of 20% H2O2, in 50℃under heat preservation decoloring 20min, then add three times the size of the corresponding concentration of ethanol, centrifugal sedimentation must take preliminary purify Morus nigra Linn polysaccharide.Polysaccharides by glucan Sephadex G-200 gel column chromatography purification, get a kind of purification polysaccharide. By TLC was opened, gel column chromatography, screw light measurement and uv absorption spectra appraisal polysaccharide purity. Purification polysaccharides have four monosaccharide composition:rhamnose, glucose, galactose and mannose.This research is to determine the components of the essential oil extracted by the water steern distillation and to analyses it GC/MS. Analysis results show that the Morus nigra Linn volatile chemical composition is very complicated, volatile organic compounds of palmitic acid, (E)-2-Hexen-1-ol, 4,6-Di-tert-butylphenol,L(+)-2,3-butanediol content are higher.The compounds were isolated by extraction, partition and column chromatography on macmpomus resin and silica gel. Chemical structures of compounds were identified by IR,1H-NMR,13C-NMR, and GC-MS method. Nine compounds were isolated and identified asβ-sitosterol(Ⅰ),salicylic acid (Ⅱ),β-sitosterol palmitate(Ⅲ),9-octadecenoic acid-ethyl ester(Ⅳ),Ethyl Linoleate (Ⅴ),undecane(Ⅵ),Octadecane (Ⅶ),Palmitic acid ethyl ester (Ⅷ),methyl linoleate (Ⅸ)Using the superoxide radical system, system of 2,2-Diphenyl-1-picryhydrazy(DPPH·), and hydroxyl radical system, the antioxidant activities of extracts from the Morus nigra Linn is studied.The results showed polysaccharide and different polarities parts extract from the Morus nigra Linn had strong radical scavenging activity towards superoxide radical system, DPPH-system and hydroxyl radical system, and with increasing concentrations of polysaccharide, its antioxidant activities gradually.
