In this paper, it first made a brief introduction to the principle of Raman scattering phenomenon and surface enhanced Raman scattering (SERS), then the Raman spectroscopy technology in biology application is given. The second chapter, melamine is used to be the probe molecules, from heating mode of agitation, heating time and chemicals ratio three aspects study the of negatively silver sol which has the best effect of surface enhanced Raman scattering (SERS), and getting better negatively silver sol. Experimental results show the best silver sol manufacture craft is that:Boiling method, MAgNO3:MNa3C6H5O7ยท2H2O=1:0.55 (1=0.53mol), heating 40 minutes. The silver sol of this method can detect substance which is 10-14g/ml concentration. The third chapter, application of new nano silver sol detects the Raman spectra of female hormones, hydrangeas DNA, camellia enzyme and human saliva, gets satisfactory surface enhanced Raman spectra of these substances. The silver sol after optimization of has very good enhancement effect for estrogen. C18H24O2 has strong Raman characteristic peaks in 865cm-1,1031cm-1,1075cm-1, 1257cm-1 and 1432cm-1. C18H24O3 has strong Raman characteristic peaks in 831cm-1,1045cm-1,1111cm-1,1278cm-1 and 1417cm-1. C18H2oO2 has strong Raman characteristic peaks in 724cm-1,822cm-1,908cm-1,1141cm-1,1254cm-1,1554cm-1 and 1925cm-1. The silver sol after optimization of has very good enhancement effect for hydrangeas DNA. Hydrangeas DNA has strong Raman characteristic peaks in 635cm-1.719 cm-1,1014cm-1 and 1205cm-1. The silver sol after optimization of has very good enhancement effect for camellia enzyme. Camellia enzyme has strong Raman characteristic peaks in 809cm-1.840cm-1.970cm-1.1149cm-1.1210cm-1. 1322cm-1 and 1451cm-1. The silver sol after optimization of has very good enhancement effect for human saliva. Human saliva has strong Raman characteristic peaks in 495cm-1.635cm-1.811cm-1.879cm-1.1001cm-1.1049cm-1. 1131cm-1.1197cm-1.1315cm-1,1561cm-1 and 1644cm-1.Through the analyzes to the several biomolecule, further verifys reliability of the new silver sol in Raman enhancement effect, while also further prove it is feasibility that surface enhanced Raman scattering (SERS) technology application in the biological sciences.
