Dynamic anxial compression(DAC) preparative chromatography technology is a kind of fast and efficient separation purification technology. With the development of separation of nature procudts and traditional Chinese medicine, the establishment and application method development of dynamic axial compression chromatography technology has been the hotspot of the separation and analysis technology.This dissertation takes the DAC-50 and DAC-80 as the investigative object, with 10μm C18 as packing material, optimize the packing method and develop the evaluation method. The packing method of DAC preparation chromatographic was established at room temperature, with ethanol as slurry solvent and the ration was 1:2(v:v). Column length was 250 mm with packing pressure of 15MPa (DAC-50) and 12MPa (DAC-80).The method of DAC column performance evaluation was developed at romm temperature, with ethanol as mobile phase at a flow rate of 55mL/min (DAC-50) and 140 mL/min (DAC-80). The detection wavelength was set at 254nm with 0.1 mL (DAC-50) and 1.0mL (DAC-80) sample load volume.According to the principle of linear magnifying technology, the hops extracts and capsaicinoids effects preative process was established by a gradual enlargement from analysis (ID4.6mm) to semi-preparation (ID20mm), preparation (ID50mm,DAC). The hops extracts eluent was evaporated under vacuum, extracted by SPE and dried after the nitrogen blowing to get the yellow solid products. It showed that the purity of hops extracts target products was more than 95% by analytical HPLC technology and the total sample recovery rate was 50.2%. The purity of capsaicinoids eluent was more than 96%.The results show that the packing methods of DAC column which was set up was simple and setTab, and the evaluation methods for column characteristics were reasonable and credible. The preparation method in this thesis for hops extracts and capsaicinoids is simple and the purity of the products is high. The application in this way will guide DAC chromatographic system promotion and application in the near future. |
