In this dissertation, polystyrene-supported organoselenium reagents and the preparation of a series of activity Vitamin D3analogues through solid-phase organic synthesis were investigated.Firstly, the intermediate of calcipotriol was prepared successfully through solid-phase synthesis. We take Vitamin D2as our raw materials and polystyrene-supported organaselenium reagents as our reaction medium. And after addition of C10-19with the polystyrene-supported organoselenium reagents, ozonation, protection, elimination and2,3-Sigma rearrangement,we get the key intermediate of calcipotriol as the target. It is worth mentioning that polystyrene-supported organoselenium reagents are used not only as a coupling agent, but also a protecting group of the most active double bond(C-10,19)at the end. As a result, the steps of the reaction can be shorten mostly and we can get the target with high de values of1α-hydroxyl in our reaction.Secondly, the preparation of some analogues of Vitamin D3through solid-phase synthesis was investigated. Vitamin D2was reacted with polystyrene-supported organoselenium reagents, and then we obtained some analogues of Vitamin D3through side chain derivatization and2,3-Sigma rearrangement reaction.And now the research is ongoing.The evident advantages of the method for preparation of the above compounds are mild reaction conditions, easy operation, high stereoselectivity, good yields, and high purities of crude products without further isolation and purification.
