Background:How to stop bleeding quickly and effectively is a pressing problem in our daily life. During surgeries, effective means of hemostasis allows a major surgery to be carried out smoothly, improving the success rate of surgery and reducing surgery complications. On the battlefield, it is one of the most effective ways to reduce soldier casualties.At present, there are already a lot of hemostatic dressings. However, allergic reactions, poor degradability and local tissue damage provide obstacles to their further application. Thus, it is of great significance to develop new types of hemostatic dressing with more effective and safer properties.Cassava starch is a kind of polymers found in plants in nature. It can be degraded by a-amylase in human body. Because of its low protein content (less than1%), Cassava starch can be used as ideal biomedical materials.The sponge-like materials have a lot of pores inside. They possess very good ability to absorpt water and swell. Oxygen and carbon dioxide can almost go through it freely, creating an excellent enviroment for the healing of wounds.Content:In this study, gelatinized cassava starch sponges were prepared by freeze-drying method. Single-factor experiment was carried out to find the relationship between different parameters, including pre-freezing temperature, pre-freezing duration and the proportion between starch and water, and water absorption of starch sponge. Orthogonal experiment was carried out to find the optimal preparation process. Platelet aggregation, platelet adhesion and in vitro dynamic coagulation experiments were carried to evaluate the hemostatic efficiency of gelatinized cassava starch sponge; cell compatibility test, hemolysis test and in vitro degradation test were carried out to estimate the biocompatibility of gelatinized cassava starch sponge.Results:the optical preparation conditions were outlined as follow:the quality ratio of starch and water was1:30; the freezing temperature was-60℃, and the freezing time was12h. The sponge prepared under the optical condition possesses three-dimensional porous architecture, whose bulk density was0.0363g/cm3, with an absorbency of13times. Results of platelet aggregation, platelet adhesion and in vitro dynamic coagulation the experiments all showed that the gelatinized cassava sponges possess a better ability to stop bleeding compared with commercially available gelatin sponge; More importantly, the results of cytotoxicity, hemolysis and in vitro degradation experiments demonstrated that the sponges have good biocompatibility, which is in accordance with the demands of National Standards GB/T16886.Conclusion:In the study, the prepared gelatinized cassava starch sponge has good hemostatic effect and biocompatibility. It has the great potential to be developed as an efficient and safe starch-based hemostatic material.
