Laccase (E.C.1.10.3.2) is a kind of containing copper oxidoreductase, with the aid of oxygen will hydroquinone (of hydroquinone) oxidation in pairs of benzene quinone polyphenoloxidase one kind, also known as hydroquinone oxidase, generally exist in fungi and plants.Because has the high stability and a wide range of substrate specificity, it in the toxic substances in the degradation of biological sensors, the construction of the industry, the fields of very good application. To deal with phenolic pollutants, as a biological catalyst, fungi paint application prospect enzyme is very good. And because the paint enzyme thermal stability, it in wastewater treatment, and functional polymer synthesis fields is quite unique application value also concerned.This article mainly discuss how to cultivate excellent white-rot fungus in the process of enzyme and paint. From first white-rot fungus strains optimization and then paint the fermentation of the enzyme and its utilization, and lignin as substrate with sediment in the influence of the enzyme to paint. The experimental results, the use of white-rot fungus liquid fermentative paint enzyme, best C/N is20, enzyme can live to4391.91IU/mL. The best training for28℃temperature, pH value of4.5best.Use alkaline substances in the treatment of the corn straw of sediment yield to get lignin paint enzyme, found that enzyme production effect than no lignin the effects of sediment is better. Wave bottles of experiment, sediment concentration of lignin7g/L, detected production paint the enzyme activity for6990.6IU/mL, other conditions remain unchanged and not add lignin sediment, paint the enzyme activity in1400.1IU/mL. In addition, add a certain amount of glucose solution can make paint the enzyme, the increase in production, data show that the glucose concentration10g/L, paint the enzyme activity in7303.6IU/mL. Track glucose addition way experimental process found that if glucose to join enzyme production of the partial liquid, enzyme live also can get improved obviously.The enzyme to paint in decoloring oxidation degradation indigo and golden orange Ⅱ was studied, and the data from experiment we get:paint enzyme degradation in the oxidation of the most suitable for when indigo pH=4.5,30℃is the best temperature, with3.9IU/mL of enzyme degradation to paint discoloring reaction, when420min is completely bleaching, can reach90%; And paint enzyme degradation decoloring gold orange Ⅱ oxidation of suitable conditions is pH=4.0,30℃, the concentration of13.5IU/mL of lacquer enzyme treatment contains gold orangeⅡ solution, reaction of500min, decoloring is completely, can achieve90%above. This paper mainly studies the enzyme how to paint in environmental protection and printing and dyeing wastewater treatment play out the positive and important meaning.
