Auricularia auricula-judae is a gelatinous basidiomycetes with higher edible and medicinal value and one of china's traditional export commodities. The traditional wood cultivation was gradually replaced by substitute cultivation.At present strain of A.auricula-judae degradate seriously and the yield and quality decreased slightly,but A. auricula-judae research has focused on cultivation technique and Auricularia polysaccharides extraction, selecting excellent A. auricula-judae strain was seldomly researched and reported.This experiment maked A. auricula-judae strains Au916 and Danpian 5 as parent materials,collected spores, isolated and identified the monocaryon, adopted Monokaryon-Monokaryon crossing, Dikaryon-Monokaryon crossing and Multi-spore crossing,identified hybrids by antagonism with parents and fluorescent staining microscopy clamp connection.Hybrids was screened through measuring mycelial growth rate of culture dish and compost, extracellular laccase activity and the resistance of the T. viride.Gray correlation analysis method was used to evaluate agronomic traits of hybrids.In the tested 224 hybrids, the mycelial growth rate of 56 hybrids in the CYM medium was significantly higher than the two parents; the mycelial growth rate of 68 hybrids in the compost was significantly higher than the two parents,laccase activity of 58 hybrids was significantly higher than the two parents. Resistance determination of A. auricula-judae hybrids to T. vider showed that 12 hybrids and T. vider confrontationly cultivante and significant antagonistic line was formed at their borderline;the inhibition rate of T. vider to 8 A. auricula-judae hybrids mycelial growth was significantly lower than the two parents; inhibition rate of 7 A. auricula-judae hybrids to T. vider mycelial growth was significantly higher than the two parentsAccording to the above experiment results,15 Monokaryon-Monokaryon hybrids,10 Dikaryon-Monokaryon hybrids,26 Multi-spore hybrids were determined to carriy on the autumn substitute cultivation.fruit body thickness, fruit body length, dry weight per sawdust bag, fruit body psychrometric ratio,green weight per fruit body, mycelial growth rate on sawdust bag, the formation rate of fruit body bud, contamination rate of sawdust bag these 8 agronomic traits were investigated emphasisly and comprehensively evalued using gray correlation analysis method. The results showed that correlation rank of 6 Monokaryon-Monokaryon hybrids (D18, D2, D5, D19, D27, D7),4 Dikaryon-Monokaryon hybrids (S23, S22, S1, S9) and 9 Multi-spore hybrids (Du 10, Du 9, Du 2, Du 51, Du 53, Du 16, Du 49, Du 1, Du 3) were ahead of the two parents, and had better agronomic trait and fruit body trait,in which the general character of D19,S22,S23,Du 2,Du 9,Du 10 outweighed other strains,and their yield surpassed parent strain over 10 percent, can be cultivated for further screening.
