13kind of Polysaccharides from Atractylodes macrocephala Koidz were obtained through hot water extracting and Ultrasonic assisted extracting, their antioxidant activity in vitro is determided.After separation and purification, determided effects of molecular weight and monosaccharide composition with ultrasonic frequency and power. The highest antioxidant activity of polysaccharide was chosen to determide its structure, solution behavior and morphology. It is modified with chemical method then determide its biological activity changes. The main contents and results are as follows:1.Polysaccharides from Atractylodes macrocephala Koidz were obtained through hot water extracting and Ultrasonic assisted extracting. Results showed:Maximum rate of polysaccharides Atractylodes is8.017%.when the ultrasonic frequency80kHz power220W which is higher than hot water extraction.2Atractylodes polysaccharides have functions of scavenging free radicals and reducing power, scavenging effects on DPPH·and·OH were especially significant, as the concentration increases, Atractylodes polysaccharide clearance rate of·OH is rising, with the ultrasonic power increases, clearance rate of·OH enhanced but scavenging effect on O-2·was relatively weak. when the ultrasonic frequency135kHz power220W which has highest antioxidative activity.3.13kinds of polysaccharide from Atractylodes macrocephala Koidz were separated and purificated,then were determinated its molecular weight and monosaccharide composition with effects of ultrasonic frequency and power. Results showed:polysaccharide from Atractylodes macrocephala Koidz extracted with hot water has greater molecular weight than ultrasonic assisted extraction, and the greater ultrasonic power is the smaller molecular weight.4. Structure of UAM was determided by chemical and instruments methods. Results showed: The molecular weight of UAM was2921, is made up of glucose, mainly connected withβ-D-1â†'3and β-D-1â†'3,6, and has certain repeated branching structure, branched with glucose at the C6position of glucose. 5. The solution behavior and morphology of UAM shows that it is molecular chain with the branch, has" porous" structure, random coil chain is existed in the solution, is easy to be influenced by external environment change. X-diffraction shows that UAM is presented with crystalline and amorphous and" two-phase" has significant difference.6. After chemically modified biological activity of UAM has improved significantly, all of sulfation, carboxy methylation, phosphorylation can improve the antioxidant ability and on glycosidase inhibitory activity, carboxymethylation has comparable inhibitory activity with positive contras, has the development potential for hypoglycemic drugs.
