Objective:To establish the model of metabolic syndrome by SD rats with the character of hypertension, disorder of glucose metabolic and lipid metabolism, obesity simultaneously. To detect concentration of AngⅡin the fat tissue of all the models. To investigate the correlation between the concentration of AngⅡand the risk factors of the cardiovascular disease and MS in FSC and FC groups, just like fasting insulin(FIN), fasting blood glucose(FBG), HOMA—IR, cholesterol(TC), high density lipoprotein(HDL), low density lipoprotein(LDL), triglyceride(TG), the weight of the rats and the weight of the fat tissue, and discuss the relationship between the concentration of AngⅡand the cardiovascular disease and MS.Methods:36 weaned rats, divided into 3 groups randomly. Three different diet were given to the three different groups, one is the normal diet for a group with 12 rats; one is high fat diet for a group with 12 rats; the other is high fat and salt diet for a group with 12 rats. After 4 weeks feeding, the weight, length, abdominal circumference, blood pressure (BP), fasting blood glucose and insulin were measured. Then measure the glucose and insulin again at the 30min,120min after having the sugar. The lipid profile like HDL, LDL, TG, and TC were detected. Weight the liver and the fat tissue. To find out whether these indexes in the three different groups have statistical significance, then evaluated whether the model of the metabolic syndrome was established successfully. After that, the concentration of the AngⅡin the fat tissue were measured by ELISA, calculate the data and find out if there is statistical significance among the three groups. To investigate the correlation between the AngⅡand the associate factors.Results:1. The abdominal circumference, weight, systolic blood pressure(SBP), diastolic blood pressure(DBP), the weight of the fat tissue, the concentration of the TG, TC, LDL, the concentration of glucose and insulin in OGTT and insulin—releasing test in the FSC group were all higher than those in the NC group, but the HDL was lower; they have statistical significance(P<0.05); the color of the liver become shadow. 2. The weight, abdominal circumference, the weight of the fat tissue, SBP, TG, TC, Omin glucose, Omin and 120min insulin in FC group were higher than those in NC group, the HDL was lower, they have statistical significance(P<0.05); the DBP, 30min insulin, LDL,30min and 120min glucose don't have statistical significance between the FC group and the NC group(P>0.05).3. The concentration of the AngⅡof the fat tissue in the FSC group, FC group were higher than those in the NC group (P<0.05). The level of AngⅡin FSC group was higher than that in FC group (P<0.05).4.The concentration of the AngⅡwas significantly correlated with LDL, FIN, TC, TG, HDL, the weight of the fat tissue and body in the FSC group(P<0.05); the concentration of the AngⅡin the FC group was significantly correlated with TC, LDL, TG, the weight of fat tissue and body(p<0.05).Conclusion:1. After 4 weeks feeding by high fat and salt diet, the rats in FSC group showed the characters of hypertension, obesity, high fasting blood glucose, high TG, low HDL. These characters were similar with MS of children. It means the young rat model of metabolic syndrome was established successfully. After 4 weeks feeding by high fat diet, the rats in FC group did not show hypertension, disorder of glucose metabolic and insulin resistance. So the characters of FC group were not similar with MS.2. The concentration of the AngⅡin the FSC and FC group increased obviously, which means AngⅡparticipate in the obesity and MS; the concentration of the AngⅡbetween the FSC group and FC group also have statistical significance, it show that AngⅡparticipate in the MS process also through the other ways besides obesity.3. The level of AngⅡin the FC group was correlated with TC, LDL, TG, the weight of the fat tissue and body; it indicated that the concentration of AngⅡwas correlated with obesity and disorder of lipid metabolic. The level of AngⅡin the FSC group was correlated with TC, LDL, TG, FIN, the weight of the fat tissue and body; it indicated that the concentration of AngⅡwas correlated with cardiovascular disease and MS.
