| The Taxol compounds and Quercetin compounds have good chemical,biological activity and strong pharmacological activity, the Taxol has good clinical curative effect to Breast cancer,Ovarian cancer and Lung cancer, and it has become a remarkable new anticancer drug; Quercetin and its complexes have many pharmacological effects: anticancer,diminishing inflammation,scavenging free radicals,immune resurrection and sterilizationk, and so on; Organic germanium complexes have many kinds of efficacy, such as regulating the body immunity and hematopoiesis functionary,eliminating the free radical,resistance of mutation,anti-cancer,anti-inflammatory,antiseptic, and so on, it has become a new human biological effects regulator and the balance adjustment drugs inside body, Its application prospect is immense. The aim of this paper is to research the course and the basic mechanism of these drugs with high application value playing efficacy. This paper is carried out from the following three aspects:1. The interaction and its mechanism between anti-cancer drug Taxol and BSA have been Studied by the methods of fluorescence spectrometry and UV-Via absorption spectrometry, and the reactions relevant kinetics and thermodynamics constants have been determined. Experimental results demonstrat there is a static combination between BSA and Taxol, and it was confirmed that the main type of the interaction is the electrostatic force. All explained that the BSA has the functions of Storage and transportation to Taxol.2. Synthesize new type complex Quercetin Germanium in accordance with the literature related and test the products by infrared spectrometry and UV-Via absorption spectrometry. And by using fluorescence spectrometry, UV-Via absorption spectrometry study the interaction and its mechanism between the complexes and BSA, and the reactions relevant kinetics and thermodynamics constants have been determined. Contrast them to the Quercetin. The results show that the complexes effectively combine to BSA with electrostatic forces, and the binding force is stronger to Quercetin itself, it is obvious that BSA has the stronger storage and transportation function to Quercetin Germanium than to Quercetin.3. Study the interaction and related mechanism between Quercetin Germanium complex and DNA, and have a speculation on combination mechanism of them by the qualitative analysis on ultraviolet absorption spectrometry and quantitative calculation on fluorescence spectrometry. And use Quercetin as probes to design experiments, contrast combining ability of Quercetin Germanium complex to DNA to Quercetin itself, according to probe principle (similar to the displacement response in chemical reaction).The results indicate that Quercetin Germanium complex combine to DNA with insert force, which prevents DNA replication and other biological activities of DNA, and what'more maybe effectively prevent the spread of cancer cells. Thus it is expected that new complex Quercetin Germanium may have certain cancer-fighting property. And the binding force to DNA is stronger to Quercetin itself, indicating that the cancer-fighting ability of Quercetin Germanium may be superior to Quercetin itself. |
