The purpose of this study was to provide ideas and approaches for selection of dosage forms, screening of prescriptions, optimizatin of techniques and evaluation of quality, by utilizing the models of biopharmaceutics and pharmacokinetics. Oral administration is the most commonly used clinical administration. Absorption of a drug is the key factor for it to be effective. Therefore, this study mainly focused on the process of drug absorption and the promoter of absorption.Literature researshes showed that salvianolic acids had a low oral bioavailability, so water-soluble components named salvianolic acids were used as object drugs. In this study, HPLC methods were established and evaluated, which determined the concentrations of rosmarinic acid, salvianolic acid B, salvianolic acid A in the perfusate and rat plasma, Models including rat in situ perfusion and kinetic research in vivo were employed systemically to investigate absorption mechanism of rosmarinic acid, salvianolic acid B and salvianolic acid A. We further investigated the effect of borneol on the absorption and pharmacokinetic of salvianolic acids.The results showed that the transport mechanisms of rosmarinic acid and salvianolic acid B were similar to active transport or facilitated diffusion. However, salvianolic acid A was passive diffusion. In vivo, salvianolic acids demonstrated a rapid distribution and eliminated rapidly from the systemic circulation. And salvianolic acids could undergo enterohepatic circulation. Borneol could enhance the intestinal absorption of salvianolic acids. The capacity of enhancing effect was considered to be significant in different intestinal sites.When concentration of borneol increased, the enhancing effect of borneol for intestinal absorption of salvianolic acids increased. While the effect decreased significantly when borneol increased to some degree. The enhancing effect of middle concentration of borneol was the optimum. Borneol also could enhance the intestinal absorption and inhibit the metabolism of salvianolic acids. In this study, absorption mechanism of salvianolic acids and the effect of borneol were investigated by multi-biological models from multi-angles, to explore the theoretical basis to improve the bioavailability and promote the absorption. The accomplishment of the project will provide a novel idea and method of researching and developing modern preparation of TCM with high effect, high quality and high standard.
