| Dipsaci Radix is the dried root of Dipsacus asper Wall, ex Henry. Dipsaci Radix, Wine-processed Dipsaci Radix, and salt-processed Dipsaci Radix are the three kinds of decoction pieces and ordinarily used in treating different diseases for their different pharmacological effects. But the processing parameters are not definitely noted in Chinese Pharmacopeia(2010,â… ), and methods of the quality control are nearly the same for Dipsaci Radix, wine-processed Dipsaci Radix, and salt-processed Dipsaci Radix except the description for their appearances and identification for cross-section.Our processing complied with general principles recorded in Chinese pharmacopeia and both wine-processed Dipsaci Radixes and slat-processed Dipsaci Radixes were processed according to the principles at different conditions, in which different amount of wine were added (from 10% to 20%), different heating temperature (from 140~150℃to 170~180℃) and heating period(from 20min to 30min) were adopted. Powders of decoction pieces were extracted by n-Bu-OH saturated with water, and then the samples were detected by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). TLC assay was carried out in silica gel G plate with upper solution of the mixture of Bu-OH:H2O:CH3COOH:CH3OH=4:5:1:0.1, with 10% sulfuric acid alcohol solution as colorimetric reagent. HPLC detection was carried out by an Alltech C18 column (4.6mm×250mm,5μm) with the mobile phase consisting of methanol (A) and water((0.5%CH3COOH, B) in gradient model, with an evaporative light scattering detector(ELSD) adopted. Chromatograms of TLC and HPLC showed that the behavior of chromatography was not significantly different among three kinds of Dipsaci Radixes pieces, but the content of several components were different. In order to continue TLC and HPLC investigation,6 kinds of wine-processed Dipsaci Radixes and 2 kinds of salt-processed Dipsaci Radixes were made for several batches with Dipsaci Radix pieces purchased from Bozhou Herb Medicine Factory. Although neither new substance generated nor substance missed, the contents of 9 kinds of chemical substances had changed. |
