HPLC Determination Of Single Index Component And Multi-Index Components In Tradition Chinese Preparation

During the research of quality standard and quality test, it is important to chose the active ingredient as index composition to establish the determination method of HPLC. High Performance Liquid Chromatography has been a important way for quality control, which has the advantages of column efficiency, high sensitivity, separation speed, good reproducibility and easy operation. In this thesis, wu studied the HPLC determination of active compositon in sanhuang tablets and ershiwuweidtang capsules.(1) Ershiwuweidatang pill is gathersed in Tibetan ministerial standard. It contains safflower, myrobalan, Mao Terminalia, and so on. It is used to appetizer, ulcer, bleeding, stomach and liver pain, loss of appetite, menorrhagia and epiataxis embolism.Ershiwuweidatang capsule comes from changing in dosage form of ershiwuweidatang pill. It's quality standard in Tibetan ministerial standard just has the TLC of Costas and Drynaria. There is no determination. In this thesis, we established a method for the determination of HSYAA in Ershiwuweidatang capsule. The conditions of HPLC and the preparation of samples were examined in detail. The results of methodology authentication of linearity, precision, stability, reproducibility and recoveries showed the method is reliable and reproducibility. Then the method was used in the stability experiment of Ershiwuweidatang capsule for determination of HSYAA.(2) Sanhuang tablets is a commonly traditional medicine. It's first gathers is in Jinkuiyaoyue, and now is gathered in Pharmacopoeia of the People's Republic of China (2010 edition). Sanhuang tablets can be used to purging heat, cooling blood, red eyes and thrat, nose and mouth sores, sore throat, sore gμms, upset thirsty, urine and constipation. In addition, it can be used for acute gastroenteritis and dysentery. And the recent pharmacological studies have shown that sanhuang tablets also can be used to anti-inflammatory, antibacterial, treatment of diabetes and high blood lipids, treatment of gynecological diseases, and so on.In this thesis we established a method for the simutraneous determination of gallic acid, sennoside A, berberine, baicalin, aloe emodin, emodin and chrysophanol by HPLC. In this study, the conditions of HPLC, such as components and gradient elution of mobile phase, detection wavelength, flow rate and colμmn temperature were examined in detail. The preparation of samples including the extraction method, extraction solvent and extraction time were examined carefully. The results of methodology authentication of linearity, precision, stability, reproducibility and recoveries showed the method is reliable and reproducibility. Then the method was applied for the determination of sanhuang tablets from different manufacturers. And the result showed that the content of the seven kinds of components from different manufacturers was different, and the most was baicalin, berberine was the smallest.In this thesis, we established the HPLC determination of ershiwuweidatang capule and sanhuang tablets, which help to enhance and improve the quality control of sanhuang tablets and ershiwuweidatang capule.
