Part One: Studies on fermentation of a endophytic fungi Shiraia sp. Slf-14 from Huperzia serrata and its componentsHuperzia serrata , also known as help- king, melaleuca tower and SheZu grass, belongs to Lycopodium Head , Huperziaceae , Huperzia Is plant and is herbaceous perennial ferns and a kind of traditional Chinese herbal medicine. The studies show that there are a lot of endophytic fungi in H. serrata and Slf - 14 fermentation can produce Huperzine A or other alkaloids, which have reversible, high efficiency, high selective inhibitory activity to acetylcholinesterase. The fermentation conditions: Culture medium is PD, Training volume is 150mL, Incubation temperature is 28℃and Training rate is 120 r/min. mycelia and fermented liquid are extracted respectively with petroleum ether, chloroform and ethyl acetate in order to get petroleum ether extracting part, chloroform extracting part and ethyl acetate extracting part, using dithio-bis (ATCh) as the substrate and glucosinolate and - 2 - nitro benzoic acid (DTNB) as colour-display reagent, acetylcholinesterase inhibitory activity of extraction parts of mycelia strains and fermented liquids is detected. The results show that acetylcholinesterase inhibitory activity of chloroform extracting part of mycelia strains is the highest. Participated by chloroform and detected in 313nm wavelength, using ultraviolet spectrophotometry can detect total alkaloid content of mycelia strains.To further clarify mechanism and material basis of acetylcholinesterase inhibitory activity of the mycelia strains, This study uses ethanol extraction, multiple distribution and silica gel RP-column chromatography or other kinds of column chromatography and studis on multiple-purification and structure elucidation of chloroform extracting part are done, we can get main chemical ingredients. This study firstly screens and researches the different organic solvent extracting parts of mycelia and fermented liquid produced by fermentation by means of acetylcholinesterase inhibitory activity so as to determine inhibitory activity of chloroform extracting part of mycelia strains is the highest and doing research on chemical compositions of mycelial chloroform extracting part for detailed clarification provides a preliminary theoretical basis for material foundation and function mechanism of acetylcholinesterase inhibitory activity.Part Two:Studies on the components of flavonoid glycosides from Arborescens ArtemisiaArborescens artemisia (Artemisia selengensis Turcz), is a perennial herb artemisia compositae, the plant tissue is rich in flavonoids, which can reduce myocardial oxygen consumption, coronary and cerebral vascular flow to increase, lower blood glucose and lipids and have physical activity of a clear action such as a clear body of superoxide anion radical, anti-aging and enhancing immunity.In order to further clarify chemical structures of flavonoid glycosides compounds of arborescens artemisia on the poyang lake, purification and structural identification of flavonoid glycosides compounds in the wild arborescens artemisia are studied , chemicals in the arborescens artemisia are extracted by means of ultrasonic auxiliary solvent extraction, and separation of flavonoid aglycone and flavonoid glycosides compounds in the arborescens artemisia by polyamide column chromatography and separation and purification of flavonoid glycosides monomer compounds in the arborescens artemisia by Sephadex LH - 20 gel column chromatography are made, according to the physical and chemical properties and the spectral characteristics of chemical structure, identification of compounds are done. Research shows that separation of flavonoid aglycone and flavonoid glycosides compounds of arborescens artemisia can be better, and separation of the flavonoid glycosides monomer compounds is established respectively in accordance with the name of chrysoeriol-7-O-β-D-glucose and phloretin-3-O-β-D-xylose.This experiment using ultrasonic auxiliary solvent extraction and combining with polyamide column chromatography can fully extract and effectively separate flavonoid aglycone and flavonoid glycosides compounds of arborescens artemisia, using sephadex LH-20 gel column chromatography can separate and prepare flavonoid glycosides monomer compounds, flavonoid glycosides monomer compounds are used as a related product quality controlling standard for natural plant arborescens artemisia, the study of the chemical composition and physiological activities lay material foundation and theoretical basis.
