| Obsjective:To explore the expression of JNK1 and TRAF2 in adipose tissue of women with and without polycystic ovary syndrome (PCOS). To analyze the relationships of JNK1 and TRAF2 to body mass index(BMI),the levels of serum sexual hormones,fasting plasma glucose (FPG,fasting insulin (FINS,insulin resistance(IR). To explore the fuction of JNK1 and TRAF2 in PCOS.Methods:Samples PCOS(Obesity n=13,Non-obese n=15)and controls (Obesity n=20,Non-obese n=20).1. follicle stimulating hormone (FSH),luteinizing hormone(LH),estradiol(E2),testosterone(T),prolaction(PRL)and fasing insulin level (FINS)were measured by chemiluminescence ansay. Fasting plasma glucose (FPG)were measured by glucose oxidase method.2. Insulin resistance index was calculated using homeostasis model assessment insulin resistance(HOMA-IR).3. Gene expression of JNK1 and TRAF2 were detected by reverse transcriptase polymerase chain reaction (RT-PCR)in the adipose tissues of the four groups.4. The relationships of JNKland TRAF2 to body mass index(BMI),the levels of serum sexual hormones,fasting plasma (FPG),fasing insulin level (FINS),insulin resistance index(IR)were analyzed by correlation and regression.Results:1. There was significant up-regulation of JNK1mRNA in adipose tissue in women with obese PCOS when compared with obese control, there was also significant up-regulation of JNK1mRNA in adipose tissue in women with Non-obese PCOS when compared with Non-obese control, JNK1mRNA was highlier expresssed in adipose tissue in women with obese PCOS compared with Non-obese PCOS.2. There was significant up-regulation of TRAF2mRNA in adipose tissue in women with obese PCOS when compared with obese control, there was also significant up-regulation of TRAF2mRNA in adipose tissue in women with Non-obese PCOS when compared with Non-obese control, TRAF2mRNA was highier expressed in adipose tissue in women with obese PCOS compared with Non-obese PCOS.3. LH,LH/FSH,T,FINS and HOMA-IR were also obviously higher in women with obese PCOS and Non-obese PCOS. There was significant up-regulation of FINS,HOMA-IR in women with obese PCOS when compared with Non-obese PCOS.4. JNK1mRNA expression in adipose tissue was positively correlated with BMI (r=0.356,P=0.006),T(r=0.639,P=0.009),FINS (r=0.561,P=0.000),HOMA-IR (r=0.491,P=0.000),TRAF2mRNA expression (r=0.652,P=0.008). TRAF2mRNA expression in adipose tissue was also positively correlated with T(r=0.556,P=0.000),FINS (r=0.626,P=0.009),HOMA-IR (r=0.548,P=0.015),JNK1mRNA expression (r=0.625,P=0.023),LH (r=0.639,P=0.005).5. Multiple regression analysis showed that HOMA-IR was related factor in influencing JNK1 and TRAF2mRNA expression.Conclusion:1. Hyperinsulinemia,hyperandrogenism and insulin resistance exist in PCOS, insulin levels and insulin resistance were higher in women with obese PCOS when compared with Non-obese PCOS. It was inferred that obesity can aggravate insulin resistance in PCOS.2. The expression of JNK1,TRAF2 in the adipose tissue are up-regulation in women with PCOS. It was inferred that it is the pathogenesis factors of the PCOS.3. The expression of JNK1,TRAF2 are related to insulin resistance. It was inferred that they are involve occurrence and development of insulin resistance in women with PCOS. |
