The Effects Of Hydrogen Sulfide In Rat Hepatic Stellate Cells On Collagen â…  And Collagen â…¢

Objective:To explore the effects of hydrogen sulfide in rat hepatic stellate cells on collagenⅠand collagenⅢ.Methods :The subcultured rat hepatic stellate cells(HSC-T6) were divided into 7 groups:blank group(group B , HSC-T6),Sodium Hydrosulfide group(to separate three groups:group N1:group B+20μmol/LNaHS ;group N2:group B+100μmol/LNaHS; group N3:group B+200μmol/LNaHS ) and Propargylglycine (PPG) group(to separate three groups,group P1:group B﹢7×10﹣5mol/LPPG; group P2:group B﹢2×10﹣4mol/LPPG; group P3:group B﹢7×10﹣4mol/LPPG).After the different concentration sodium hydrosulfide (NaHS,donor of H2S) and propargylglycine(PPG,a metabolic enzyme inhibitor of H2S) were given,collagenⅠand collagenⅢcontent in supernatants were Determined by ELISA.To explore the effects of hydrogen sulfide in rat hepatic stellate cells on collagenⅠand collagenⅢ,different concentrations of exogenous H2S donor sodium hydrosulfide and different concentrations of H2S formation of endogenous enzyme cystathionine-γ-lyase inhibitor propargylglycine intervene HSC.At 6h, 12h and 24h time points,collagenⅠand collagenⅢcontent in supernatantswere determined by ELISA.To explore H2S in rat hepatic stellate cell collagenⅠandⅢcollagen expression.Results:In the control group (normal cell group) collagenⅠat 6h and 12h was no significant difference between,but there were significant differences compared with 24h.CollagenⅢat 6h were significant differences compared with 12 and 24h,However, no significant difference between 12h and 24h.Under normal culture of hepatic stellate cells was significantly high expression of collagenⅠandⅢcollagen.After adding sodium hydrosulfide hepatic stellate cells and secretion of collagenⅠandⅢcollagen decreased.After giving PPG,collagenⅠand typeⅢcollagen expression was significantly increased.Conclusions:Normally a large number of activated hepatic stellate cells and collagen expression of collagenⅠandⅢ,administration of exogenous H2S donor NaHS treatment,the supernatant of collagenⅠand typeⅢcollagen decreased.H2S formation of endogenous enzymes to give cystathionine-γ-lyase inhibitor of PPG,the supernatant of collagenⅠand typeⅢcollagen content increased.That indicate that hydrogen sulfide can inhibit rat hepatic stellate cells and collagenⅠandⅢcollagen expression and secretion and inhibit the development of liver fibrosis.
