Background:The gastric cancer is one of the most common tumor in Chine, serious harm to human health.Surgery is the main treatment, but because the majority of patients with varying degrees of the existence of micro-metastasis, postoperative recurrence and metastasis in patients with advanced gastric cancer is often clinically, so chemotherapy in the comprehensive treatment of gastric cancer in a very important position. With the development of molecular biology and clinical application of individualized attention, optimize the treatment plan, realize the basic research and drug individualized become common concern for clinical trials.Research mainly is in of biomarkers detection, predict the therapeutic effect and prognosis, guiding drug,molecular targeted drug research and clinical application. EGFR, Her-2/neu were already been confirmed in a wide variety of tumor tissues, and cancer prognosis has relevance, and as a molecular targets guidance molecular targeted drug application. TopoⅡalpha is many chemotherapy drugs such as Teniposide, Adriamycin Etoposide, etc, the identification of drug targets. Its content can be determined by the variety of tumor sensitivity or resistance to anticancer drugs target.Objective:1.To investigate the expression of EGFR,Her-2/neu,TopoⅡαin gastric cancers and their relation-ship with clinical pathologic character. Explore theirs clinical significance as the molecular targets to guide the molecular targeted therapy of gastric.2.To investigate the feasibility of EGFR,Her-2/neu and TopoⅡαas theraptic target of molecular targeted therapy of gastric cancer? and to provide a theoretical basis to therapy and evaluation of prognosisMethods:1.To collect 108 cases of gastric adenocarcinoma and 22 cases of normal gastric tissue with follow-up and clinicopathologic data.2.To detect EGFR,Her-2/neu and TopoⅡαexpression in gastric cancer and normal gastric tissue by immunohistochemistry assay.3.To statistically analyze the relationship between EGFR,Her-2/neu and TopoⅡαexpression and various clinicopathologic parameters using SPSS 17.0.Results:1.The 41.67%,31.5%,53.7%, respectively of 108 gastric cancer specimens for EGFR,Her-2/neu and TopoⅡαwere positive, Normal gastric tissues were positive and 0%,0%,4.5%.There are the significant relation between two groups(P<0.05).2. The expressions of EGFR,Her-2/neu and TopoⅡαwere not correlated with sex, age, size (P>0.05).The positive expression rates of EGFR,Her-2/neu and TopoⅡαwere related to the site of gastric cancer, cell differentiation, invasion depth, lymph node metastasis and TNM staging (P<0.05).3.There are significantly positive correlationship between EGFR expression and Her-2/neu expression in gastric cancer, it is also between EGFR and TopoⅡα,between TopoⅡαand Her-2/neu. Conclusion:1.The positive rate of EGFR, Her-2/neu and TopoⅡαexpression in gastric cancer is higher, compared with EGFR, Her-2/neu and TopoⅡαnegative expression in begin gastric disease.2. EGFR, Her-2/neu and TopoⅡαexpression was not correlated with sex, age and size and was related to site of tumor and degree of differentiation, depth of invasion, clinical stage,lymph node and distant metastasis and survival of gastric cancer patients.The results suggested that the cases with EGFR, Her-2/neu and TopoⅡαexpressive were prone to local invasion, lymph node and distant metastasis.3. There are signifieantly positive correlationship bewteen EGFR expression and Her-2/neu expression in gastric cancer,It is also with bewteen EGFR expression and TopoⅡαexpression in gastric cancer,bewteen Her-2/neu and TopoⅡα.
