| Ocular trauma is the major cause of impairment of vision, especially it's the cause of monocular deprivation. Traumatic optic neuropathy (TON), as a common disease, always results in secondary or primary loss of vision, optic nerve is injured not oniy by brain or orbit injury indirectly, but also direct optic nerve axotomy. There is a complicated pathology process after optic nerve injured. Multiple factor influence the reparative regeneration of injured optic nerve, which is the only way to cure traumatic optic neuropathy. So finding effective device and mechanism to promote the regeneration of optic nerve has become a focus in the world.In empirical study of the regeneration of optic nerve after injury, the effect of lens injury which leads to cataract is approved gradually, meanwhile many domestic and overseas scholars pay close attention to its mechanism. Ciliary neurotrophic factor and leukemia inhibitory factor are the members of interleukin 6 family, vitro and animal studies have improved exogenous CNTF/LIF can promoted the regeneration of retina ganglion cells, but there is rarely report whether endogenous CNTF/LIF take part in the process of lens injury promoting the regeneration of injured optic nerve, we make correlated exploration by animal study.ObjectiveOur research is to investigate the effect of CNTF/LIF on the regeneration of injured optic nerve with lens injury, by detecting the expression of CNTF/LIF in retina. MethodFifty-six healthy adult Wistar rats without disease of the eyes are divided into 3 groups at random:normal control group (8 rats) without making any dea, simple optic nerve axotomy group (24 rats)(completely cut optic nerves at 3mm behind the eyeball) and joint injury group (24 rats)(optic nerve axotomy beside lens receives intraocular penetration). Normal control group rats are extirpated eyeballs after 7 days of feeding; 7,14 and 21 days after operation,8 eyeballs are extirpated respectively in other 2 groups. The histopathological changes of retina and RGCs are observed by HE staining. GAP-43, CNTF and LIF are labeled in retina by immunohistochemistry and measured by Biosens Digital Imaging System v1.6 system, the data are made statistical analysis.Results1. HE staining:Every level of the normal rat retina is clear; In simple optic nerve axotomy group, the thickness of retina decrease more serious than joint injury group and the number of RGCs is much less than joint injury group.2. GAP-43 immunohistochemistry staining:In normal rat retina there is no expression of GAP-43, and the integral optical density(IOD) is 59.49±0.99/rat. Simple optic nerve axotomy group, IOD of 7 day group is 80.98±1.62/rat; IOD of 14 day group and 21 day group are 60.44±1.29/rat and 59.34±1.32/rat, they are both loss than that of 7 day group, they have significant statistical differences (P=0.000), but There are no significant differences statistically between 14 day group and normal group,21 day group and normal group(P=0.118; P=0.784). In joint injury group, IOD of 7 day group is 121.70±1.99/rat, IOD of 14 day group and 21 day group are 122.94±1.19/rat and 122.20±1.0/rat; There is significant difference statistically between each time point and normal group(P=0.000), but There are no significant differences statistically among 7 day group,14 day group and 21 day group (P=0.26).7 days after surgery, There are significant differences statistically among each treatment group(P=0.000); 14 days and 21 days after surgery, There are significant differences statistically among each other treatment group(P=0.000) except normal group and Simple optic nerve axotomy group.3. CNTF/LIF immunohistochemistry staining:there has little expression of CNTF/LIF in normal rats, and the integral optical density(IOD) is 70.25±1.14/rat and 68.55±0.68/rat. Simple optic nerve axotomy group, IOD of 7 day group is 84.63±0.87/rat, there is significant difference statistically between 7 day group and normal group(P=0.000); IOD of 14 day group is 61.33±0.66/rat, less than 7 day group and normal group, their differences are significant statistically(P=0.000). In joint injury group, IOD of 7 day group and 14 day group are 121.48±0.87/rat and 121.30±0.71/rat, both more than that of normal group, their differences are significant statistically(P=0.000), but here is no significant difference statistically between 7 day group and 14 day group((P=0.667).7 day and 14 day after surgery, there are significant differences statistically among three treatment groups(P=0.000).Conclusion1. Lens injury could everely increase the expression of GAP-43 and promote the regeneration of retina ganglial cells after optic nerve axotomy.2. Lens injury could significantly promote the expression of Endogenous CNTF, and the expression state is stable.3. Endogenous CNTF/LIF take part in the process of Lens injury promote the regeneration of retina ganglial cells after optic nerve injury. |
