1.Objective:To observe type 2 diabetic rats combined sepsis of pancreatic oxidative stress in mitochondria,the APS was used in type 2 diabetic rat combined sepsis,and to elucidate Protective effect of APS on Oxidative Stress of pancreatic mitochondria injury in type 2 diabetic rats with sepsis.2.Methods:Forty-five rats were randomly divided into normal control group,sham CLP group(DM group),CLP1 (saline group),CLP2 (low dose group) and CLP3 (high dose group).High fat diet for 5 weeks,type 2 diabetic rats were established by abdominal injection of a small dose (30mg/kg) of streptozotocin(STZ). After the modeling, the use of Astragalus polysaccharides pretreatment 1 week,and then septic model was copied by cecal ligation and puncture (CLP).The activities of Na+K+-ATPase,Ca2+Mg2+-ATPase and total ATPase,superoxide dismutase(SOD)and glutathion peroxidase(GSH-Px) the contents of malondialdehyde(MDA) and nitric oxide(NO)in the panereatic mitochondria were observed respectively.3.Results:The activities of Na+K+-ATPase,Ca2+Mg2+-ATPase,total ATPase,SOD and GSH-Px in the panereatic mitochondria in the high-dose and low-dose group were increased more obviously than the saline group(P<0.05),while the contents of MDA and NO in the pancreatic mitochondria in the high-dose and low-dose group were decreased obviously (P<0.05).The contents of MDA and NO in the panereatic mitochondria in the saline group were increased more obviously than the normal control group(P<0.05),while The activities of Na+K+-ATPase, Ca2+Mg2+-ATPase,total SOD and GSH-Px in the panereatic mitochondria in the saline group were decreased more obviously(P<0.05).4.Conclusion:2 diabetic rat combined sepsis have been badly damaged of pancreatic mitochondrial by oxidative stress,APS can effectively reduce the injury of Oxidative Stress of pancreatic mitochondria in type 2 diabetic rats combined sepsis.5.Significance:It is chosen for Astragalus polysaccharides for the active ingredients of Astragalus mongholicus frequently used in Traditional Chinese Medicine in the topic.In vivo studies,we observe type 2 diabetic rats combined sepsis of pancreatic oxidative stress in mitochondria,the APS was used in type 2 diabetic rat combined sepsis,and we elucidate Protective effect of APS on Oxidative Stress of pancreatic mitochondria injury in type 2 diabetic rats with sepsis and its mechanism.It is a scientific basis to provide for the clinical application of Traditional Chinese Medicine.
