Objective:Established the animal model of transection lesion of spinal cord,in order to observe the effect of body weight support treadmill training (BWSTT) and electric acupuncture in changing the GAP-43 and Caspase-3 in the spinal cord, and study the primary mechanism on the transection lesion of Spinal Cord.It will provide the theory evidence for the experimentally cure the transection lesion of spinal cord.Methods:There are 65 healthy female Wistar adult rats(clean grade),weight is 180-220g, are divided into 5 group: normal group(5 rats), sham-operated(15 rats), model control group(15rats), BWSTT and electric acupuncture group (15 rats), electric acupuncture group(15 rats).Each group is further divided into3d,7d,14d,21d,28d time groups, every group have 5 rats, after medical experimental dispose, that deal with BBB scale and immunohistochemistry protocol. The data that are obtained from the animal model, is the expressions of GAP-43 and Caspase-3, by the SPSS 14.0 statistical analysis.Results:The walking function obstacle is evident when the operation, transection lesion of spinal cord rats model, is successful. The expression of GAP-43 in the healthy adult rats of spinal cord was inexistence. But the expression of GAP-43 was increased in the spinal cord after the spinal cord injury by the operation. The expression of GAP-43 in the spinal cord after injury was increased at day 3, and further increased to the highest expression at 7day, then the expression of GAP-43 was reduced steadily,but the data is more than normal. The expression of Caspase-3 in the transaction lesion of spinal cord was rare. But the expression of Caspase-3 was increased in the spinal cord after the spinal cord injury by the operation. The expression of Caspase-3 in the spinal cord after injury was increased at 3day,and further increased to the highest expression after seven days, then the expression of Caspase-3 was reduced steadily,but the data is more than normal. Three days after the surgery,the data obtained is not obvious difference, but 7 days later, all of the data show that three groups have evident difference (p<0.05) at the same time.Conclusion:1. BWSTT and electric acupuncture that can increase the data of GAP-43 in the most significant, can reduce the transection lesion of spinal cord rats model that damaged pathomorphology.2. BWSTT and electric acupuncture that can decrease the data of Caspase-3 in the most significant, can reduce the transection lesion of spinal cord rats model that damaged pathomorphology.3. BWSTT and electric acupuncture can increase the data of BBB scale in the most significant.
