| Objective:To change of the light environment conditions on the sleep of wild type Drosophila.Methods:Experiment 1 Different intensities light impact on the sleep of fruit flies.In this study,CantonS strain of Drosophila is used as observation,we collected unmated Drosophila eclosion within 8h,and divided into four groups and estimated 20001ux,40001ux,60001ux,80001ux.we will move the Drosophila to different intensities light control box that is 20001ux,40001ux,60001ux,80001ux right now.and feeding for 6 days.The experiment began to monitor at 7:00.Drosophila fruit flies sleep are defined as the standard still longer than 5min.In this experiment,fruit flies sleep time and activities are used to index.The effects of different intensities light on the 24h of sleep of Drosophila and sleep-wake rhythmExperiment 2 Changing of light intensity on the seventh day impacts on the sleep of 7 days DrosophilaIn this study,CantonS strain of Drosophila is used as observation,we collected unmated Drosophila eclosion within 8h,and divided into five groups, and rear in the basic training for 6 days. We calculate the sleep time and sleep-wake rhythm in DrosophilaExperiment 4 Continuous light deprivation impacts on the sleep of 7 days DrosophilaIn this study, CantonS strain of Drosophila is used as observation,we collected unmated Drosophila eclosion within 8h,and divided into two groups and rear in the basic training for 7 days. And placed the Drosoplia on the DAMS in the 7 days morning.We will move each group to the light control box before 19:00 in the evening.Every evening,8:00 begain to light and continue light for 10min,the whole point turn on the light for 10min and continuous monitoring for several days.Control group is monitoring in the basic environment,and experiment is end when a group of Drosophila are all died. We calculate the sleep time and sleep-wake rhythm in DrosophilaExperiment 5 Extension time of the light continuously impact to sleep of fruit flies.In this study, CantonS strain of Drosophila is used as observation,we collected unmated Drosophila eclosion within 8h,and divided into four groups and rear in the basic training for 7 days. And placed the Drosoplia on the DAMS in the 7 days morning. We will move each group to the light control box that prolonged illumination 1h,2h,3h before 19:00 in the evening Control group is monitoring in the basic environment, monitoring began to7:00 at eight morning,and 24h continuous monitoring. We calculate the sleep time and sleep-wake rhythm in Drosophila.Result:Experiment 1:Different intensities light impact on the sleep of fruit flies during the day.Male and female with the increase in light intensity during the day,night and total sleep time has increase.①Day:male fruit sleep time is most during 40001ux and female fruit sleep time is most during 60001ux.②ight: light intensity on the male fruit files sleep is nothing impact,thus,female fruit flies'sleep time during 80001ux are most at night.③Total sleep time :male fruit flies'sleep most when the light intensity is 80001ux,and female'sleep most when the light intensity is 80001uxExperiment 2 Changing of light intensity on the seventh day impacts on the sleep of 7 days DrosophilaWith the increase of light intensity,male and female flies'sleep time are increasing during night and total time.①Day:during the day,with the light intensity increasingly,male flies'sleep time decreased,and 80001ux is lest.thus,female flies'sleep time don't change significantly.②Night:male and female fruit flies'sleep time are most when the light intensity is 20001ux.③Total sleep time:the result of male and female are contrary,male flies'sleep time are least when the light intensity is 80001ux,thus,female flies'sleep time are most when the light intensity is 20001ux.Experiment 3 Extended time of light out impacted on sleep of 7 days DrosophilaMale and female flies in alternating light and dark different time,fruit flies'sleep time at night have increased,comparing with control group.Thus,sleep time during the day is reduced,comparing with control group, total sleep time compared with control group don't change significantlyExperiment 4 Continuous light deprivation impacts on the sleep of 7 days DrosophilaGiven the light stimulation to male and female flies will cause sleep time increasing at night and sleep of the next day showed a rebound phenomenon.But,male and female flies don't reduce sleep time with the increase of days of intermittent light stimulationExperiment 5 Extension time of the light continuously impact to sleep of fruit fliesCompare with control group,male and female flies'sleep time don't change significantly with the extension photoperiod.However,male and female fruit flies during the day,night and total sleep time are increasing with the extension of light time.Conclusion:Experiment 1:Different intensities light impact on the sleep of fruit flies during the day.Drosophila selective light differently, fruit flies'sleep time are different under rear in different light intensityExperiment 2 Changing of light intensity on the seventh day impacts on the sleep of 7 days DrosophilaChanging of 7 days Drosophila's light intensity could cause sleep time change.
