Purpose: To establish the optimal extraction process of compatibility of Radix Aconiti Lateralis Praeparata (RALP) with Glycyrrhiza and that of Radix Aconiti Lateralis Praeparata with Rhizoma Zingiberis (RZ) in"Ginseng Sini Decoction"; to study the changes of active substances content in their compatibility.Method: 1. the optimal extraction process of compatibility of RALP with Glycyrrhiza and that of RALP and RZ was established by using L9(34) orthogonal experiment method 2. the active substances content changes in compatibility were determinated by HPLC and acid-base titration.Result: 1. The optimal extraction process of compatibility of Glycyrrhiza with RALP: for extracting twice, water added for 10 times, extracting time 2 h, and the concentration of the alcohol for precipitate is 70%. 2. The optimal extraction process of compatibility of RZ with RALP: for extracting third, water added for 20 times, extracting time 0.5 h. 3. Most diester diterpene alkaloids of RALP, i.e., toxic ingredients, were hydrolyzed after decocting. 4. After compatibility, the content of glycyrrhizin, liquiritin, and the total alkaloids were obviously reduced, comparing with those of separate decoctions of Glycyrrhiza and RALP. The total alkaloids of combinational decoction is obviously higher than that of separate decoction of RALP, however, the content of the 6-gingerol is decreased after their compatibility.Conclusion: In the aspect of the changes of compatibility of RALP and Glycyrrhiza, the composition of Glycyrrhiza (Glycyrrhizin, Liquiritin) accelerates the hydrolysis of toxic diester diterpene alkaloids into cardiac ester alkaloids, and Glycyrrhiza plays a role in detoxification in"Ginseng Sini Decoction". What's more, the combination of RALP with RZ makes the content of the total alkaloids increased. It shows that the constituents of RZ promote the dissolution of aconite alkaloids to increase the content of active substances in combinational decoction. It offers synergy effects. The changes prove the effects of the incompatibility of RALP and Glycyrrhiza, and the compatibility of RALP and RZ. It provides scientific basis for clinical application and medicie exploiture, and establish foundation for the further research.
