OBJECTIVE:Safflor yellow (SY) is one of the active constituents of traditional Chinese medicine safflower. It comprises various chalcones chemical compound, existing in the water-solubility part of safflower. It has been proved that SY can remedy the focal cerebral ischemic reperfusion injury of heart and brain, including reduce infarct area, improve the activity of Orgotein(Superoxide Dismutase,SOD) of cardiac muscle or blood plasma, obliterate oxygen radical and calcium antagonists, improve microcirculation, reduce the water content,the number of calcium ions, glutamic acid, aspartic acid, glycine and malondialdehyde in the cerebral hemisphere of I/R rats. However the function mechanism of SY is still not clear. We attempt to discuss the effects of SY on the I/R in rats in this study.METHODS:72 health adult male Wistar rats (SPF) were divided into 4 groups randomly, that is:sham operation, model control, low-dose group(2.5mg/kg), and high-dose group(5.0mg/kg). Middle cerebral artery occlusion (MCAO) was used to prepare focal cerebral ischemia model. The reperfusion happened 1h after the insert of the line. The rats of the SYL group and SYH group were injected with SY before the insert of the line and instantly after the insert of the line. Sham operation and model control group were injected with same volume of physiological saline. Rectal temperature, ECG, Blood pressure, and rCBF on each rat were detected before MCAO,0.5h during and after 2h. Using the hrv model of powerlab the ECG was studied getting the frequency domain indexes. After the detection the effects of the drug were studied about neurological deficit, the water content of brain tissue, the cerebral infarcted zone, and the change of pathology under microscopic examination.RESULTS:Compared to sham operation, the rats of the model control group neurological deficit, the edema of brain tissue, the cerebral infarcted zone were observed obviously. The SYL and SYH group were much remissive in the above aspects comparing to the control group.The rCBF of the sham operation kept stable. The rCBF of model control, SYL, and SYH decreased obviously after the insert of the line, indicating success of the model.2h after the reperfusion the rCBF of the latter 3 groups regained. But rCBF of the SYL and SYH were much bigger than the model control, witch indicating SY could increase the rCBF after the reperfusion.The MBP of the model control increased obviously after the insert of the line. 2h after the reperfusion the MBP decreased slightly. The MBP of the SYL and SYH decreased more obviously comparing to model control after the reperfusion, coming nearer with sham operation.The high power (HF) of frequency domain about hrv of the model control decreased obviously after the insert of the line, and increased after the reperfusion. The SYL and SYH increased more obviously than the model control. The total power (TP) decreased obviously after the insert of the line, and increased after the reperfusion. The TP of the SYL and SYH increased more obviously than the model control.The change of MAP and hrv showed that SY could reduce the damage of autonomic nerves system.CONCLUSIONS:SY can reduce the neurological deficit, the water content of brain tissue, the percentage of infarction area and the pathological change in the cerebral hemisphere.SY can increase rCBF after the reperfusion, improve the blood supply of the ischemia area.SY can improve the function of the autonomic nerves system which is damaged by the ischemia. It can reversion or lighten the dysfunction of the autonomic nerves system after the ischemia.
