Objective:8-Prenylnaringenin (8-PN) is the most powerful phytoestrogen by far. It was reported that 8-PN can improve the experimental postmenopausal osteoporosis in rats. However, it is still unknown whether its function due to the effect on the bone formation or bone resorption. The present study was to observe the effects of 8-PN on nuclear factor NF-κBligand(RANKL), osteoprotegerin(OPG), and macrophage colonystimulating factor(M-CSF) of mice osteoblast cell line (MC3T3-E1), and with different genistein, daizein and other plant estrogen compared to effect intensity. The other objective was to find the change that 8-PN that treat on osteoclast of the number and the area of resorption pits, and with different genistein, daizein and other plant estrogen compared to effect intensity.Methods:Different concentrations of 8-PN(10-8,10-7,10-6 M), genistein(10-6 M), daizein(10-6 M) were used to treat on RAW264.7 with M-CSF and RANKL as induction agents for eleven days. find the change of the osteoclast's number and the area of resorption pits. Different concentrations of 8-PN(10-8,10-7,10-6 M), genistein(10-6M), daizein(10-6M) were used to treat on MC3T3-E1 for six days, use the method of RT-PCR to test change of the RANKL, OPG, M-CSF of the sample's mRNA, and use the method of ELISA to test the RANKL and OPG of the sample's protein density. Estrogen (10-8 M) was used as positive control in all the experimental groups.Results:8-PN can decrease the osteoclasts number and the area of resorption pits with dosage dependence. The effect is weaker than E2 and stronger than other plant estrogen.8-PN can restrain the expression of RANKL and promote the expression of OPG with dosage dependence, has little change of M-CSF. The ratio of RANKL/OPG was decrease. The regulate of the ratio is weaker than E2 and stronger than other plant estrogen.Conclusion:8-PN can restrain the differentiation and bone resorption in osteoclasts. The effect is stronger than other plant estrogen. And can regulate and control the expression of RANKL and OPG in MC3T3-E1 to indirect regulate and control the osteoclasts.
