Objective:To observe the effect of bezafibrate on the levels of fasting blood glucose (FBG), fasting insulins(FINS), triglyceride(TG), free fat acid(FFA) in the blood,and the change of expression levels of PPARγin hepatic tissue of the type 2 diabetic rats induced by high suger and fat feed, with low dose streptozotocin(STZ).And explore the possible mechanism of reduce insulin resistance of type 2 diabetic rats in hepatic tissue.Method:(1)sixty SD rats were randomly divided into normal control group (NC, n=15) and diabetes group(DM, n=45).The NC rats were fed with regular chow and DM rats were fed with high suger and fat diet. After 8 weeks,the DM rats were injected with 0.5% STZ 30mg·kg-1 once to make SD rats of type 2 diabetes models. Thirty type 2 diabetes models were formed sucessfully in total,then the models were randomly divided into 2 groups by number table method:diabetic control group (DM-C) and diabetic group treated with bezafibrate (DM-B),with 15 rats in each group. DM-B rats received intragastric administration of bezafibrate(50mg·kg-1·d-1),while NC rats and DM-C rats were given equal doses of solvent for 12 weeks. The body weight were examined,the levels of Fasting blood glucose (FBG),Fasting insulins (FINS), triglyceride(TG), Free fat acid (FFA) et al were detected before and after intervention. Homeostasis model assessment for insulin resistance (HOMA-IR)were calculated.(2)After 12 weeks intervention,all rats were sacrificed.The PPARy expression in hepatic tissue were detected by immunohistochemistry.Results:(1)At the end of experiment,body weight,FINS in DM-C and DM-B group were lower than NC group(P<0.01),while FBG were higher than NC group (P<0.01). (2)After 12 weeks intervention,the body weight of DM-B group were up-regulated compared with DM-C group (P<0.05), while the levels of FBG,FFA and TG were decreased markedly (P<0.01).Compared with before intervention,the levels of FBG,FFA,TG,FINS and HOMA-IR in DM-B group were improved at the different degree.(3)At the end of experiment,the expressions of PPARγof DM-C group decreased markedly compared with NC group (P<0.01), while the expressions of PPARγof DM-B group were similar with NC group(P>0.05)Conclusion:Bezafibrate decreased FBG, FFA, TGand reduce the lipotoxicity, and improve insulin sensitivity of type 2 diabetic rats in hepatic tissue. Which might be related to the up-regulated expressions of PPARγin hepatic tissue.
