| ObjectiveTo investigate the influence of MSCs transplantation on premature ovarian failure induced by 4-vinylcyclohexene diepoxide (VCD).MethodsFemale C57 mice were dosed with VCD (160mg kg-1 day-1, ip) daily for 20 days to induce ovarian failure. Estrous cyclicity was monitored by daily vaginal cytology at 8 every morning. Ovarian failure was assigned when mice showed 10 days or more of persistent diestrus.Animals were randomly assigned to two groups: VCD-treated group and vehicle control group. The VCD-treated group was divded into two subset, the one for MSCs transplantation and the others use PBS as substitution.GFP-labeled BMSCs were injected into the ovary in 100th after the onseting dosing in VCD-treated mice. The level of FSH and LH in the serum were detected at 2w, 4w and 6w after transpantation. the ovaries were collected in 6w after transpantatipn. Part of fresh tissues were use to make ice-cold slash for GFP-labeled MSCs tracking in the ovaries. Marker expressed on the surface of steroidogenic cells of ovaries were also detected by immunofluorescence.Part of ovaries were kept in -80℃for total RNA extraction.ResultsPathological section shows no follicles can be seen obviously in VCD-treated group and the level of FSH and LH were higher than vehicle control.The concentration of FSH and LH in MSCs transplant group reduce from 4w after transplantation.Green fluorescent cells were tracked in ovarian tissues of every time point after transplantation, and FSHR and LHR that expressed in steroidogenic cell can not be detected in those cells.The RT-PCR showed lower FSHR and LHCGR in MSCs group than that in PBS group.ConclusionsMSCs can migrate in the mouse ovaries and exist in ovarian tissues of mouse for a long time, but cannot differentiate into steroidogenic cell in ovaries. The result from Serology, pathology and RT-PCR indicated bone Marrow Stem Cell Transplantation produce a false image as repairmen on mouse injured ovarian induced by VCD.In fact, it seems a kind of ovarian failure progression. The underlying mechanism might be MSCs maldifferentiate into other cells. |
