The purpose of the test was to study the relationship between obesity and gut microflora, and the effect of Bacillus on gut microflora of obese rats. Firstly, the obesity model was established through feeding rats high-fat diet in order to study the effect of obesity on gut microflora. Secondly, the suspension of Bacillus was given to obese rats, At the fourth and seventh week of Bacillus treatment test, gut microflora of obesity and obesity resistant rats, short chain fatty acids(SCFA) of fecal and rats'weight were tested.The main test results were as follows:1. The technique of denaturing gradient gel electrophoresis(DGGE) was used to analyze the changes of total microflora structure. The results showed that the gut microflora diversities of obese rats were significantly higher than that of control rats, and there were new dominant microfloras produced in gut of obese rats, but there were other dominant microflora gradually disappeared. Compare with obese rats, the gut microflora diversities of obesity Bacillus rats were significantly higher, but that of obesity resistant rats were lower, and all the differences were obvious. In addition, the weights of obesity Bacillus and obesity resistant rats were lower than that of obese rats.2. The technique of ERIC-PCR was used to analyze the change of Enterococcus. The results showed that the change of Enterococcus kept the same pace with that of gut total microflora. The differences between all groups were obvious. Especially, the Enterococcus diversities of obesity resistant rats were very low(P<0.01).3. The technique of gas chromatography(GC) was used to analyze the SCFA in the rats'feces. The results were that the SCFA content of obesity Bacillus group was the highest, that of obesity group was second, and that of obesity resistant group was the lowest. The change of Anaerobic bacteria could be reflected by the results of SCFA content indirectly, the Anaerobic bacteria change of obesity and obesity Bacillus groups kept the same pace with gut total microflora, but the Anaerobic bacteria quantities of obesity resistant rats were low relatively.All the test results showed that there was direct relation between obesity and gut microflora, obesity made gut microflora change greatly, and there were new dominant microfloras produced; The gut microflora of obesity animal could be improved by Bacillus, the main result was that increasing the gut micro flora diversity of obese animals, and Bacillus could play a positive role in weight control of obese animal.
