Objective: To explore the promoting role of carcinoma-associated fibroblasts on human breast cancer cells through SDF-1/CXCR4 signaling axisMaterials and Methods:1.primary culture of CAFs and NAFs :8 breast cancer tissue blocks and 8 normal breast tissue blocks were obtained from 8 patients who were confirmed to suffer from breast infiltrating ductal carcinoma by pahthological section over the period of May 2010 to August 2010. 2.Quantitative RT-PCR Quantitative real time PCR was performed to measure the SDF-1 mRNA levels of the CAFs and NAFs ,also the CXCR4 mRNA levels of MCF-7 and 231 breast cancer cell lines. 3.Three-Dimensional Ago-Gel Cell Culture System :control group1(MCF-7 cells only) ;control group2(MCF-7+NAFs);controlgroup3(231only); experimentalgroup1(MCF-7+CAFs);experimentalgroup2(MCF-7+anti-SDF-1Ab+CAFs;experimentalgroup3(231S+CAFs).Results:Compared with normal stromal fibroblasts ( NAFs ) ,breast carcinoma associated fibroblasts(CAFs) displayed distinct SDF-1 gene expression and MCF-7 cell line had elevated expression of CXCR4 mRNA compared to MDA-MB-231 Scell line .In the three dimension agarose system,CAFs showed to promote the cloning of human breast cancer cell lines compared to NAFs.Anti-sdf-1 monoantibody also could relatively inhibit the interaction between CAFs and human breast cancer cell lines.Conclusion:the expression of SDF-1 in the carcinoma- associated fibroblasts(CAFs) enhances the cloning in human breast cancer cell lines .This suggests that for human breast cancer cells ,the SDF-1/CXCR4 signaling pathway may play an important role in the growth of these cells.
