Objective:To investigate IDH1 gene mutation in the development of adrenal pheochromocytoma.Methods:Using PCR and DNA, mutation of 4 exon IDH1 gene in 10 patients with adrenal pheochromocytoma was analyzed by PCR and Bi-directinal DNA sequencing analysis.Results:The IDH1 exon 4 fragment was amplified with specific primers from adrenal pheochromocytoma DNA samples (n=10), and subjected to 2% agarose gel electrophoresis. The correct PCR product was sequenced and no nucleartide mutation was found at the presumpted site; while 100% samples presented a A to G switch in the IDH1 intron.Conclusions:The mutation on IDH1 exon was not found while on IDH1 interons was detected in adrenal pheochromocytoma .The mutation of IDH1 interons but not exon may due to gene polymorphism and may not correlated with development of adrenal pheochromocytoma.
