Objective To investigate the impact of chloride channel blocker (niflumic acid, NFA) on the impression of mCLCAmRNA and MAPK of pulmonary artery smooth muscle cells in rats.Methods 24 healthy SD rats ,randomly divided into 3 groups, which are Control group,Hypoxic group,Hypoxic+drug group,each group has 8 rats. Constructed the hypoxic pulmonary hypertension rat model. The hypoxic+drug group was cultured in hypoxic box for 3 weeks, 8 hours per day, and with NFA injected intraperitoneal 3mg/kg each day. The hypoxic group was cultured with the same circumstance as the hypoxic+drug group but were injected placebo .Changes of right ventricle was tested by RVHI;changes of mCLCAmRNA in PASMCs were tested by RT-PCR; changes of MAPK were tested by SABC immunohistochemistry.Results RVHI of hypoxic group was higher than the other groups(p<0.05);expression of mCLCAmRNA was higher than the other groups(p<0.05);expression of MAPK was higher than the other groups(p<0.05).Conclusion NFA can obviously reduce the impression of mCLCAmRNA of PASMCs of HPH rats; can block-up the activation of MAPK pathway ,can reduce the hypertrophy of the right ventricle,so the NFA have the function of cure HPH.
