Objective: To study the expression of HMGB1 mRNA and protein in extrahepatic biliary epithelial cells(EBECs) after the infection of rotavirus.Methods: 1. Compared the HMGB1 protein expression in HBECs with or without RRV infection by immunohistochemical staining in vitro.2. Examined the protein and mRNA expression after viral infection with/without small interfering RNA targeting HMGB1(siHMGB1) transfection in vitro.The HBECs and supernatant were harvested at 12h,24h and 36h,48h for quantitative real-time PCR and enzyme linked immunosorbent assay(ELISA).Results: 1. HMGB1 was localized in the nucleus of HBECs.2. RRV infection decreased HMGB1 in the nucleus of HBECs.3. At 12h and 24h, the HMGB1 mRNA expression in RRV group was apparently higher than that of negative group(P<0.01)and siHMGB1 group was lower than RRV group(P<0.01).4. At 36h and 48h, the HMGB1 concentration in supernatant between RRV group and negative group were quite different(P<0.01)and siHMGB1 group was lower than RRV group significantly (P<0.01).Conclusion: 1. RRV infection increased HMGB1 mRNA expression and extracellular HMGB1 protein . concentration2. SiHMGB1 can significantly inhibit the HMGB1 concentration in supernatant.
