Female CBA/J mice were mated with male Balb/c and DBA/2 mice respectively to construct normal pregnant and spontaneous abortion models.2. The immunocytochemical assay was used to measure the expression of Tim-3 and Gal-9 in the trophoblast cells of murine RSA and normal pregnant models, and then a highly precise color-image measure system was applied to a statistical analysis. 10 visual fields of the high power lens were chosen in every case, and then the mean optic densities were calculated where Gal-9 was positive. ã€Results】1. The normal pregnant and spontaneous abortion models were successfully constructed.2. Cytoplasm of trophoblast cells both in normal pregnant and RSA models turned into brown, which suggested Tim-3 was positive in the trophoblast cells. The level of Tim-3 in RSA models was significantly lower than that in normal pregnant models(P <0.05).3. Gal-9 was positive in the trophoblast cells both of normal pregnant and RSA models. Compared to normal pregnant models, the level of Gal-9 in RSA models was significantly lower than that in normal pregnant models(P <0.05).ã€Conclusion】Tim-3 and Gal-9 were expressed in the trophoblast cells of normal pregnant and RSA models. The abnormal expression of Tim-3 and Gal-9 may result in RSA.
