Aim: To investigate the effect and mechanism of 15 - epimer - lipoxin A4 reduces reactive oxygen species generation in BV-2 microglia induced by lipopolysaccharide.Methods: Interfere with 15-epimer-lipoxin A4 to BV-2 cells in 30 minutes, and then BV-2 cells were stimulated with 100ng/ml LPS to induce inflammatory response. Use microplate reader to detection the fluorescene of ROS after stimulation with LPS at different times. p47phox protein express in cytoplasm and cell membrane were detected by western blot, and directly observed membrane translocation of p47phox protein by confocal immunofluorescene.Results: Compared with LPS group, 15-epimer-lipoxin A4 significantly reduced the generation of LPS-induced Reactive oxygen species (ROS),and inhibited the membrane translocation of p47phox protein.Conclusion: In this study we can find that 15-epimer-lipoxin A4 inhibits LPS-induced pro-inflammatory mediators, so it can block or relieve the harmful effect by the means that inhibit the development of nueroinflammation.
