AimDetect differences in the expression of multiple Wnt genes and proteins in human breast fibroblasts (CAFs) and normal breast fibroblasts (NBFs), then discuss the mechanism of CAFs abnormally activating Wnt signaling pathway of mammary epithelial cells by paracrine.Methord1. Obtain nine persons'surgical specimens whose pathological type is invasive breast cancer. Isolate CAFs and NBFs from these specimens, then cultivate these cells.2. Detect the expression of Wnt3A, Wnt5A and Wnt10B genes in CAFs and NBFs by Real-time quantitative PCR.3. Detect the expression of Wnt3A, Wnt5A and Wnt10B proteins in CAFs and NBFs by Western blot.ResultThere is no significant difference between the expression of Wnt3A gene in CAFs and NBFs (p>0.05). The expression of Wnt5A gene in CAFs was significantly lower than NBFs (p<0.05). The expression of Wnt10B gene in CAFs was significantly higher than NBFs (p<0.05). There is obvious difference between the expression of Wnt3A,Wnt5A,Wnt10B proteins in CAFs and NBFs(p<0.05). The expression of Wnt protein is significantly increased in CAFs.ConclutionCAFs can influence the Wnt signaling pathway of mammary epithelial cell by increasing the expression of Wnt3A, Wnt5A and Wnt10B proteins to promote the incidence of human breast cancer. The expression of Wnt10B gene and protein has shown a consistent increase, This shows that Wnt10B may be the perfect target for further research.
