Objective:To study the effect of the HBX protein on the expression of c-Jun through AKT signalling in liver oval cells.Methods:1. Detected the expression of HBX through fluorescence microscope, Western Blot and Cell immunofluorescence.2.Investigated the expression of p-AKT and c-Jun protien in pEGFP-HBX-LE6 oval cells through Western Blot. L02-HBX cells, pEGFP-LE6 oval cells and LE6 oval cells used as control groups.3.Bloked the PI3K/AKT signalling pathway by a specific inhibitor of PI3K/AKT signalling pathway that was called LY294002, Investigated the expression of c-Jun protien in pEGFP-HBX-LE6 oval cells through Western Blot.Results:1.The pEGFP-HBX-LE6 oval cells and pEGFP-LE6 oval cells showed green fluoresence; HBX protien was expressed in pEGFP-HBX-LE6 oval cells; Immunofluorescence indicated that HBX protien was expressed in pEGFP-HBX-LE6 oval cells and located mainly in nucleus.2.The expression of the protein level of p-AKT,Akt and c-Jun were upregulated in the pEGFP-HBX-LE6 oval cells.3.The expression of c-Jun was decreased, especially in pEGFP-HBX-LE6 oval cells after the blocking of the PI3K/AKT signalling pathway.Conclusion:HBX protien has the ability to upregulate the protein level of c-Jun and Akt signalling in oval cells; Moreover, the upregulated c-Jun protein level is dependent on the HBX-upregulated AKT signalling. |
