Objectives:To develop a method for establishment of diabetic mouse model and investigate the effects of Discolor Cinquefoil Herb extract on the levels of sugar, TC, TG, SOD, MDA and NEFA in blood of Alloxan-induced diabetic mice. Methods:Mice was induced to be diabetic by two methods:①a single high dose intraperitoneal injection of 200 mg/kg body weight (BW) of alloxan and②high-fat diet for a month followed by single intraperitoneal injection of 90 mg/kg body weight(BW)of alloxan. High-fat diet and alloxan-induced diabetic mice were used in this study and treated with extract of Discolor Cinquefoil Herb by intragastric administration. The effects of Discolor Cinquefoil Herb on the glucose tolerance and the levels of serum glucose, TC,TG,SOD,MDA and NEFA were investigated. In the study, the total Discolor Cinquefoil Herb decoction was extracted with boiling water, and the extract of Discolor Cinquefoil Herb decoction was rotary evaporated under reduced pressure at room temperature followed by drying in a freeze-dryer. Three fractions of dry extract of Discolor Cinquefoil Herb decoction were generated by cooling filtration (FractionⅠ), following extraction with 100%,50% alcohol (FractionⅡ), and remainder after extractions of 100% and 50% methanol (FractionⅢ). Then investigate the total extract of Discolor Cinquefoil Herb decoction and it's three fractions on body weight, spleen and other organs coefficient, fasting blood glucose (FBG), liver glycogen,.serum free fatty acids (FFA), total cholesterol (TC), triglyceride (TG) were measured. In order to investigate the blood glucose and lipid metabolism, determine the most effective fraction of Discolor Cinquefoil Herb and provide a theoretical basis for the further application and development of Discolor Cinquefoil Herb we make this research.Diabetes mellitus (DM), one of the most common chronic and systemic disease in the world, is characterized by hyperglycemia resulting from defects in insulin secretion, insulin action or both. It is classified into two types:type 1 (insulin-dependent) and type 2 (insulin-independent). Type 1 diabetes is caused by an absolute deficiency of insulin secretion fromβ-pancreatic cells. In the other, type 2 diabetes is closely associated with obesity and is characterized by a combination of resistance to insulin action and an inadequate compensatory insulin-secretory response.Results:1 There are two ways to make the mice to be diabetic that high-fat diet for a month followed by single intraperitoneal injection of 90 mg/kg body weight (BW) of alloxan is superior to a single high dose intraperitoneal injection of 200 mg/kg body weight (BW) of alloxan and we choose the former one.2 Discolor Cinquefoil Herb make a gradual improvement in the spirit of diabetic mice, increased activity, weight gain, improvement symptoms, improvement of the organs factor.3 Discolor Cinquefoil Herb lower diabetic mice blood glucose significantly, improve glucose tolerance, increase hepatic glycogen content. The function of the fraction II was the strongest.3 Discolor Cinquefoil Herb reduce diabetic mice serum free fatty acid significantly, total cholesterol and triglyceride levels and alleviate the abnormal lipid metabolism.4 Fraction II is most effective. It is possible that through the role of the gastrointestinal tract to regulate blood glucose metabolism, promote liver glycogen synthesis and inhibit the liver glycogen breakdown.Conclusions:In conclusion, the present study found that Discolor Cinquefoil Herb extract prevented hyperglycemia and dyslipidemia in diabetic mice. Gene expression analysis revealed that Discolor Cinquefoil Herb extract treatment affected expression level of the genes related to lipid metabolism and glycometabolism. Therefore, Discolor Cinquefoil Herb should be considered as a candidate for future studies on diabetes. In addition, further comprehensive pharmacologic investigations, including experimental long-term studies, should be carried out on Discolor Cinquefoil Herb or its bioactive compound.
