Experimental Study On Qifangweitong Against Indomethacin-induced Stomach Ulceration In Rats

Objectives: To evaluate the protective activity of Qifangweitong Granule(QFG) against the indomethacin-induced stomach ulceration in the rat model and correlates with its histological maturation,antioxidative and epithelialization .Methods: Sprague-Dawley rats were divided into four groups. Ulceration in the rats was induced with a single dose of indomethacin of 3mg/ml dissolved in distilled water. The control group rats were given the normal saline; the ranitidine group rats were given the ranitidine of 30mg/ml; the QFG low dose group were given the QFG of 1.25mg/ml; the QFG high dose group were given the QFG of 2.5mg/ml ,1ml/100g once daily by oral intubation for 7 days starting from 4 hours after the indomethacin administration. After 7 days, the stomach tissues were excised for histological examination (ulcer index,thickness of regenerative mucosa and the expression of EGFR) and the heart blood were drown for biochemical analysis (the quantity of MDA,SOD,EGF in blood serum).Results: Treatment with QFG and Ranitidine for 7 days could effectively heal the stomach ulceration as revealed from the ulcer index and thickness of regenerative mucosa. Compared to the controlled group, those treated with QFG and ranitidine reduced the ulcer index,increased the thickness of regenerative mucosa,decreased the content of MDA in the serum,increased the contents of SOD,EGF in the serum,improved the expression of EGFR in the regenerative mucosa respectively. And the effect of high dose of QFG being better.Conclusion: QFG can protect indomethacin-induced gastric ulceration revealed from the ulcer index,the thickness of regenerative mucosa and the expression of MDA,SOD,EGF,EGFR, those might be the possible mechanisms for improve the quality of ulcer healing(QOUH).
