Objectives To analyze the application value of 4-5 h-short-time in-vitro fertilization (IVF) by comparing the clinical outcome of 4-5 h-short-time IVF versus conventional IVF. Methods In the present study, all the oocytes studied were collected from the conventional control ovulation stimulating cycles, which were allocated into two groups according to two different fertilization ways given to the patients, namely 4-5 h-short-time IVF group (the testing group) and conventional IVF group(the control group). In the testing group, 90 patients were recruited and 90 cycles were performed, the oocytes were exposed to the sperm for 4-5 h before denuding. In the control group, 90 patients'oocytes of all the 90 cycles underwent the removal of the partial granular cells after the oocytes and sperm were blended together for 16h to 18h. Totally, fertilization rate, including 2PN, 1PN fertilization and 3PN, cleavage rate, normal cleavage rate, D2 and D3 high-quality embryo rate, clinical and biochemical pregnancy rate in both groups were compared and analyzed by statistical software, respectively.Results The testing group vs t the control group: the total fertilization rate (92.31% vs94.00%), normal fertilization rate (85.09% vs85.82%), abnormal fertilization rate (14.91% vs14.18%), the total cleavage rate (96.27% vs93.38%), normal cleavage rate (99.74% vs98.35%), D2 high-quality embryo rate (43.67% vs40.90%), D3 of high-quality embryo rate (27.91% vs24.09%),clinical pregnancy rate (35.56% vs32.22%) and implantation rate (23.96% vs23.33%). There are significantly statistical difference between two groups in total cleavage rate and the normal cleavage rate(P <0.01) ; There is significantly different in D3 high-quality embryo rate (P <0.05), and no statistically significant in others(P> 0.05). Conclusions In comparison of convention IVF, 4-5 h-short-time fertilization is a simple, effective, accessible way and can get the similar clinic-effect and replace conventional fertilization way.Objectives To analyze the application value of short-time in-vitro fertilization(IVF) by comparing the clinical outcome of 5-6 h-short-time IVF versus conventional IVF. There are significant difference between two groups in fertilization rate , cleavage rate, normal cleavage rate, D2 and D3 high-quality embryo rate (P <0.01); There are statistically significant between two groups in the implantation rate and clinical pregnancy Rate (P <0.05).Conclusion In comparison of conventional IVF, the clinical result which is from 5-6 h-short-time IVF is much better, and short-time IVF may replace conventional IVF. Objectives To investigate the different sperm-egg coincubation time on the results of IVF in the short-term fertilization program, and whether granular cells removal or not after fertilization on the results of IVF, to explore a safe and efficient people IVF short-term fertilization program.Methods In conventional IVF-ET cycle, totally 247 patients who got oocytes > 7 were recruited and randomly divided into four groups: A group of 44 cases of 822 eggs were collected, after adding sperm 4 - 5h, transfered into the fresh culture medium, and completely denuded all eggs'granulosa cells. B group of 1322 eggs were collected from 80 cycles, adding sperm for 5-6h and transferred into the fresh culture medium, completely removed all eggs'granulosa cells; C group, 815 eggs of 42 cycles were collected in after adding sperm for 4-5h and transfered into the fresh culture medium, without removing the granulosa cells of all eggs; D group of 1320 eggs were collected from 81cycles were transferred into the fresh culture medium after adding sperm for 5-6h, without removal of granulosa cells. Fertilization rate, 2PN rate, abnormal fertilization rate, cleavage rate, 2PN cleavage rate, high-quality embryo rate of D2 and D3, implantation rate and clinical pregnancy rate were compared among the groups and analyzed with statistics.Results Group A and B: the total fertility rate between the groups, D2 and D3 days of high-quality embryo rate, implantation rate, biochemical and clinical pregnancy rate have statistical significance (P <0.01); Group C and A: the total cleavage between the two groups in the total rate, implantation rate, D3 high-quality embryo rate was statistically significant (P <0.05); Group C and B: the total fertility rate was statistically significant (P <0.05); Group D and A: the total cleavage rate, D3 high-quality embryo rate and implantation rate have statistical significance (P <0.01), D2 high-quality embryo rate, clinical pregnancy rate was statistically significant (P <0.05); Group D and B: group B in the total fertility rate has statistically significant (P <0.01), group D in the total cleavage rate was statistically significant (P <0.05); Group D and C: no statistical significance between groups (P> 0.05) Conclusions The study shows that it can significantly increase the high quality embryo rate hand improve the pregnancy rate that removal the granular cells after human oocytes mixed with sperm for 5-6h and transferred into the fresh culture medium. This method can improve the effect of short-term fertilization in IVF program.
